Effect On Chromosome And Morphological Variation Of Populous Adenopada In Tissue Culture With Colchicine

.The shoot tips of populous adenopadaplantletswere taken as the experimental materials in this paper.With the colchicin soaking methodand co-culture method to treat material,the induction effect by different colchicin densities and the treating times had been studied.The leaf,stem,diameter,height,rooting and stomatal characteristics of materials were compared.Fitler out the appropriate treatmen of P.adenopoda screened in vitro shoot tip,to suggest possible ployoloids.The main results are as follows:1.Effect on deadth rate of populous adenopada shoot tips with colchicine.As the concentration(0.05%～0.2%)and treatment time increasing(12h～36h)the by soaking method,the death rate improved,the lowest is 15.56%,the highest reached 78.89%.As the concentration(25mg·L-1～100mg·L-1)and treatment time increasingthe(5d～15d)by co-culture method.the death rate improved,the lowest is 6.67%,the highest reached 75.56%.2.Effect on Leaf length and width of populous adenopada with colchicine.Leaves of the Variation increased and deepened.By soaking method,the best is 0.1%or 0.2%Concentration with 24h.By co-culture method,the best is 50mg·L-1 concentration with 10d or 15d.3.Effect on Stem diameter of populous adenopada with colchicine.Stem segments of the Variation thicke significantly.By colchicine soaking method,the best is 0.1%or 0.2%Concentration with 24h.By co-culture method,the best is 50mg·L-1 concentration with 10d or 15d.4.Effect on the height of populous adenopada with colchicine.Tthe Variations growth rapidly and rubustly.By colchicine soaking method,the best is 0.1%or 0.2%Concentration with 24h,.By co-culture method,the best is 50mg·L-1 concentration with10d or 15d.5.Effect on the rooting rate and rooting number with colchicine.as the concentration and treatment time increasing,rooting rate was dropping significantly and No.of rooting ruducing.Combined with effect on Morphological variation.By colchicine soaking method,the best is 0.1%Concentration with 24h.By co-culture method.the best is 50mg·L-1 concentration with10d or 15d.6..Effect on Stomatal Size and Stomatal Densit with colchicine.Stomatal of the increased,the stomatal density decreased.By colchicine soaking method,the best is 0.1%or 0.2%Concentration with 24h.By co-culture method,the best is 50mg·L-1 concentration with10d or 15d.Comprehensive analysis the effects on deadth rate,morphological variation,by colchicine soaking method,the best is 0.1%Concentration with 24h.By co-culture method.the best is 50mg·L-1 concentration with10d.The leafs is large,stems is sticker,plants is rubust and stomatal increased.The polyploids is most possibility.The research would have a guid about exploration on populous adenopada polyploidy.
