The chromosome number of Elymus dahuricus is 2n=42,and that of Hordeum brevisubulatum is 2n=28.The hybrid F1 chromosome number is 35 and the high sterility is obtained.The chromosome identification of the strong material is carried out,and some morphological characteristics are compared with the hybrid F1,the results are as follows:1.Materials F1-1,F1-2,F1-3,F1-4,F1-5 chromosome numbers are all 36,and materials F1-6,F1-7,F1-11,F1-12 are mixed-ploid plants with chromosomes of 28,36 and42.when colchicine concentration is 0.1%,the treatment time is 36h,and can be induced successfully.2.There was no significant difference in the plant height between F1 and Elymus dauri,and the material F1-11 and F1-12 were significantly higher than other materials.3.The leaf length,width and leaf thickness of the seed material should exceed F1 of the hybrid,and the leaf color will be deepened.The difference of leaf thickness is the most obvious.4.The average stomatal length,the width of the stomata and the width of the guard cells were more than the hybrid F1.The number of hybrid F1 and the number of chromosomes could be distinguished by 22 Mu long stomatal apparatus.However,because of the existence of the mixed ploid,the length of the stomata and the two kinds of materials were all intersecting,and the difference was reduced.The stomatal density of hybrids F1and chromosome 36 is about 2:1.5.the pollen fertility of F1 hybrids was only 1.14%,and the pollen fertility of F1-7was 10.56%except that of other materials.The other materials were higher than 30%.6.Hybrid F1 has a firming rate of 0,high sterility,and only 21.26%of the strong material is F1-7,with more than 50%of the rest.Pollen fertility can affect the seed setting rate.
