Study On The Vegetative Propagation Of Osmanthus Fragrans L

Osmanthus fragrans Lour., a species in the family Oleaceae, is a famous aromatous plant of Chinawith a long cultivation history, by the love of our people. The major object of this study is to exploit thetechnique systems of tissue culture and rapid propagation for Osmanthus fragrans at low cost, high survivalrate of cuttage grafting. Using the Huangchuan Jingui as the materials, which come from Henan Universitycampuses and Kaifeng Yuwang Park, in order to develop the optimum conditions for tissue culture ofOsmanthus fragrans are optimized and support for large-scale, industrialized production of Osmanthusfragrans; Using the test-tube plantlets’ leaves of Huangchuan Jingui to induce the callus and proliferationof culture；In addition, the cuttage grafting technology of Osmanthus fragrans are improved. There aremajor conclusions as follows：1. The best disinfection processing of shoot tips of Osmanthus fragrans twigs is using0.1%HgCl2dispose2.5min with survival rate up to95.6%, Alcohol produce a large damage when disinfecting the shoottips of Osmanthus fragrans, is not a good choice of tender materials sterilization.2. The most suitable initial media of shoot tips of Osmanthus fragrans twigs is B5+2.5mg/L6-BA+0.05mg/L NAA+3%sucrose, germination rates of up to86.7%.3. The most suitable regeneration medium of shoot tips of Osmanthus fragrans twigs is B5+2.0mg/L6-BA+0.05mg/L NAA+3%sucrose, K-factor up to2.08.4. The optimum rooting conditions for organization of Osmanthus fragrans culture is1/2MS+2.0mg/L NAA+full of darkness, rooting rate of up to88%, use of nutritive soil for transplanting matrix.5. Best of cytokinin in callus induction in Osmanthus fragrans leaf are zeatin, ideally inducing culturemedium is MS+1.0mg/L of zeatin+2.0mg/L IBA+3%sucrose, induction rate of up to95.6%.6. Osmanthus fragrans in the cutting process, closed without hormone treatment survival rate of95%or more, greatly reduced the production cost.In cuttings should pay attention to the bottom to sites close tothe bud, because the nutrient-rich, content of endogenous hormones, easy to root.7. In the process of grafting of Osmanthus fragrans, pay attention to the wind water, otherwise scion is easy to dry up and die due to lack of water. And the exodermis of Osmanthus fragrans is thin, so inarchingis the best choice in grafting.
