In order to better for appraisal,use and domestication of blueberry germplasm resources,this paper analysed genetic diversity of blueberry with the method of molecular marker.The paper analysed and discussed the genetic similarity degree relationships among75varieties of Vaccinium ssp,including53cultivated varieties,22bog bilberries and2lingonberries.The main results are as follows:(1)According to survey results,the wild blueberry distributes in two kinds of land:wet land and the mount areas.And their fruits include round shape,ellipse shape,pear shape and big ones.(2)The paper got a improved CTAB method for blueberry blade of surface waxy and containing polyphenol,polysaccharide and pigmentum through comparing CTAB method and plant genomic DNA extracted kit method.(3)The improved SRAP-PCR system (total20μL) was established as:.40μmol/L primer concentration;20ng template DNA;10μL2×Taq PCR Master Mix,making up the remaining volume of deionized water;Optimal annealing temperature of PCR amplification program was50℃.(4)The paper screened out15primer combinations of distinct and rich in polymorphism from80primer combinations.The genetic similarity of cultivated variety was ranged from0.5670to0.8774.The genetic similarity of cultivated variety and wild variety was ranged from0.3525to0.4751.The genetic similarity of wild blueberry was ranged from0.6207to0.9387.(5)The dendrogram of77Vaccinium samples was established using UPGMA,the result show that,77samples could be classified into three major groups in the threshold value of0.57:cultivated blueberry variety,bog bilberry and lingonberry. Indicating that the cluster results had some relativity with the pedigree of cultivars.
