Crop straw is abundant and it is the most affordable renewable energyresource in nature. High-efficient straw lignocellulose degradating microorganismscan make up the reduction of the ore energy in the world. The plentiful renewableresources-cellulosic materials could be used to produce fuel alcohol and methane bycellulose enzyme. But it is very limited for hig-cost producers of celluloseenzyme.The screen of microorganisms is critical for developing competitivebiotechnologies for biodegradation of crop straw. High-efficient straw lignocelluLose-degradating microorganisms were screened and their effects on straw degradationwere studied.By using celluLose congo-red culture medium and disintegration of filter paperto repeat screening from the wild of the Giant Panda.There were13strains which8species were fungi and5species were actinomycosis which could produce celluasewere isolated.we got one highly active celluLolytic strain W-3whose CMC enzymeactivity was2303.4U/mL(pH4.6、50℃,ug/mL·min) on carboxymethyl cellulose-Na,FPA enzyme activity was0.845U/mL(pH4.6,50℃,umol/mL·min) on filter paper.Polymerase chain reaction (PCR) was used to amplify the region of ITS fromrDNA of W-3. We got a gene fragment and sequence and which was sequenced.Phylogenetic tree of ITS gene was constructed.And the gene sequence ofrepresentative strain (CICC2487)exhibited high similarity (100%) with the ITS geneof Aspergillus niger from GenBank database.The optimum conditions for celluase-produeing were studied in this paper. theoptimized cultivation parameters of ignocellulose degradating microorganisms W-3were obtained as follows: inoculum quantity2.69×106cfu/L;5g cellulose concentration;the pH3.0-3.5; culture temperature25℃; culture time120h.Variation of CMCenzyme activity, proteins,FPA enzyme activity and the value of pH were investigated in liquid culturing process under ptimum conditions.The research indicates that anovel fungi capable of degradation cellulose was screened. we got the Aspergillusniger W-3with the activity of CMC enzyme3000U/mL.
