Anther culture of wheat is an important means of wheat haploid breeding, Providea solid foundation for wheat improvement and creation of new quality. There are manyfactors that affect wheat anther culture, Previous studies defined the ability of wheatanther culture affected by gene regulation, but because of the complexity of generegulation has not been able to get substantial progress, improve the ability of wheatanther culture is mainly concentrated in optimization of the choice of the trainingsystem and culture conditions.Study found that,Synthetic hexaploid wheat--durum wheat and Aegilops hybridcreated, with the desired resistance to disease resistance and can be effectivelytransferred to common wheat. However, the use of synthetic wheat variety improvementis very difficult, breeding efficiency is very low, In order to improve efficiency in theuse of synthetic wheat, clear the different of the resilience of anther culture withsynthetic wheat and common wheat. The important issue is whether to improve thesynthetic wheat anther culture efficiency by improve the training system, Improved theapplicability of synthetic wheat.Plant exogenous auxin2,4-D, plays an important role in plant tissue culture withembryoidid induction. It is a necessary condition to start the microspore cell division toform callus, In this experiment, different concentrations of2,4-D, with different growthstages of wheat seedlings and in different ways, in order to draw the best approach for2,4-D.The main results are as follows:1,Compared the Induction mediaC17、N6、MS with W14-F and differentiationmedia1/2MS with190-2-Cu, The results is W14-F/190-2-Cu equally suitable forvarieties of wheat in Heilongjiang Province, it is3.74times,4.67times and5.91timeswith others, Embryoidid germination rate is1.28times that of the differentiation media1/2MS, W14-F/190-2-Cu is the best system for wheat anther culture。2,suitable capacity of anther culture with synthetic hexaploid wheat is better thancommon wheat in the both sides, the embryoids of Synthetic wheat varieties averageis47.80%was37.80%of the average common wheat embryoid rate1.26times. The average survival rate of11.99%is the seeding rate of7.39%to1.62times.3, Spraying the solution of10ml/L2,4-D in the tillering stage seedlings grown inthe field, Embryoidid induction rate of anther culture Spray processing rate of thecontrol of the experimental material in embryoidnic28.00%to22.66%is1.24times.This shows that the embryoidid induction rate on wheat anther culture seedlingstillering stage, high concentrations of2,4-D spraying a role in promoting. Thedifferentiation rate is18.67%lower than the controlled trials.4, Pre-treatment period10ppm2,4-D soaking inhibition of wheat anther culture ofembryoidid induction.after2,4-D soaking the materials out of the embryoid was18.73%lower than22.66%of the untreated control group experiments in embryoidnic.Thisshows that the concentration of10ppm of2,4-D solution soaked wheat young spikes,anther culture induced embryoid rate has a certain inhibitory effect. The differentiationrate is21%lower than the controlled trials.5, Induction media supplemented with3ml/L2,4-D solution on the inhibition ofwheat anther culture of embryoidid induction.compared with the normal media2ml/Lof2,4-D test. induction media supplemented with3ml/L2,4-D.corresponding derivedembryoidid induction rate was19.96%,slightly lower than in the control test groupinduction rate of22.66%.This indicates that in1L of culture media by adding3ml/L2,4-D on anther embryoidid induction inhibited.And the differentiation rate is14.85%lower than the controlled trials.
