In this study, derivative lines were obtained from the F1 hybrids between recombinant inbred lines RIL66 and RIL181 with the blood of SHW-L1 and Qinling rye. Neat lines were selected from F4 progeny. The chromosome number and constitution of F4 derivative lines were identified by molecular cytogenetic analysis. The results were as follows:1. Ten neat lines were got from F4 progeny of RIL 66 x Qinling rye because of their g ood agronomic character. The chromosome number of these neat lines was 42. It was indic ated that the eight lines had no rye genetic material by GISH analysis. All rye chromosome s eliminated preferentially in F4 progeny, having the same chromosome and constitutions with common wheat. Two lines had rye genetic materials by GISH analysis, one is 2R/? m onosomic substitution line and another is 2RS-?translocation line by FISH analysis. Numb er 16 lines (no rye chromosome) were resistant to eight Chinese Pst races (CYR30, CY31, CY32, Sul 1-4, Sul 1-5 and Chuanmai-42) at the adult stage.2. A new 1BL.1RS translocation line A-2 was characterized in F4 progeny of RIL 181 x Qinling rye by GISH analysis. The chromosome arm 1 RS was from Qinling rye and was different from German rye Petkus. Gliadin bands of different S1 seeds of A-2 were the same with the Gld 1B3 encoded by IRS. But among these eleven individuals plants of S2 progeny, gliadin bands of different seeds of four individuals segregated. There were two phenomena. Gliadin bands of some seeds had Gld 1B3 bands encoded by IRS. Gliadin bands of some seeds varied with the loss of some gliadin bands and the loss of Gld 1B3 bands encoded by 1RS. The mutation rate was 44.44%. It was indicated that the individual plant with no Gld 1B3 bands was not 1BL.1RS translocation line with the chromosome number 42 and no rye genetic material by GISH analysis.1BL.1RS translocation line and no 1BL.1RS translocation line coexisted in S2 progeny.
