The main root of seedlings is always appropriately cut by people to generate lotsof lateral root,so it is easyly to survive in the afforestation. In this experiment,we takeMin43of the main Camellia oleifera cultivars of Fujian Province as as the researchobject.after cutting off the root tip of Camellia oleifera,we treat the wound withexogenous hormones and biotin mixture,and they have a greater influence on thewound healing and the formation of integrity lateral roots,and so on. though theanalysis of anatomical characteristics of each treatment, the determination ofphysiological indicators of each treatment,the search of root architecture of eachtreatment,we designed to explore the factors wich have effective influence on thewound healing of Camellia oleifera seedling root,so we can promote the healing ofwound rapidly to settle the contradictory between the trauma of cutting root and thenurturing of high quality seedlings. The results are as follows:1、The effect on wound healing of root cutting of Camellia oleifera with Biotinmixture treament is the most obvious, biotin mixture can complete healing in about15d, compared with the control treatment,it is16d in advance. The IAA treament took29d, compared with the control treatment, it is2d in advance.KT treament took thethe longest time to healing the wound of cutting roots,it is38d.By the observation ofanatomical characteristics of root cutting wound, the callus is formed chiefly bypericycle cells and cortical cells, the effects of the different treatment weresignificantly different: IAA treatment make cortical cells grow and elongate in11dafter the cutting root,and the cell size become larger; KT treatment make cortical cellsdivide in17d after the cutting root, the number of the cells become larger and the sizebecome smaller, the growth of the pericycle cells was inhibited. Biotin mixturetreatment make cortical cells grow in9d after the cutting root,and the callus wasformed by both cortical cells and pericycle cells in the thirteenth day of experiment.2、The role of IAA on the growth of Camellia oleifera seedling lateral root isthe mostly obvious,the tateral roots of IAA treatment appear in the the seventeenthday of the experiment.The appearances of control treatment, biotin mixture treatmentand KT treatment respectively lasted23d,27d and40d. IAA and KT play theopposite role in the induction of lateral root primordia, The plants of the two plantsshowed a huge difference,and the position of the role are all the main root of the Camellia oleifera seedlings.the new lateral roots of the plants with IAA treatmentmainly appear on the the primary root,there is no emergence of new lateral roots onthe lateral roots before cutting, while the opposite result on the emergence of newlateral roots with KT treatment. Early in the experiment,the effect of biotin mixture onthe growth of lateral roots is not obvious, but in the latter, biotin mixture treatment aresignificantly better than the control treatment on the the length or number of lateralroots.3、The fluctuations of plants with control treatment in physiological indicatorsare relatively larger, IAA treatment and biotin treatment both can alleviate the changesfrom the stress of root cutting and restore physiological indicators to normal levelwhen the lateral roots emerge. In the early stage, difference between KT treatment,and control treatment are insignificant, However, in the mid-and late,the fluctuationsin soluble protein content and SOD activity of the plants with KT treatment arerelatively larger,but the chlorophyll content remained unchanged.4、After the growth of Camellia oleifera seedlings restore to normal state, thedescending order of the topological indexs of the roots in different treatments is asfollows: IAA> Control> KT>Biotin, the topological index of IAA treatment are thelargest, and the root branching is the most simple, closely to the ningyo. thedescending order of the box dimension of the roots in different treatments is asfollows: IAA> Control> KT>Biotin, the box dimension of IAA treatment are thelargest,and its branching are the most capable.5、The biotin mixture can promote the wound of root cutting to heal rabidly,however, IAA can induce a large number of lateral roots and complete the supply ofwater and nutrients, The recovery of physiological indexes of plants with IAAtreatment are better than the ones with biotin mixture treatment. Therefore, Therefore,the protective measures to root injury should be combined with the both advantages ofthe biotin mixture and IAA,while plants complete the wound healing in the shortesttime,it also can generate a large number of lateral roots, restore normal growing.In this experiment, though the research on effective factors in the wound healingof Camellia oleifera seadings,we explore the method which make seedlings restorerapid ly to normal growth and provide people with reference methods and theoreticalbasis for the actual afforestation, This is important for improving the survival rate ofafforestation of Camellia oleifera seedlings.
