Cordyceps militaris is an important medicinal and nutritious edible fungus. Inthis study,we collected30Cordyceps militaris. Using ISSR molecular markerstechnology to investigate the difference of Cordyceps militaris strains on themolecular level and to establish the genetic map of the Cordyceps militaris toanalysis the diversity of germplasm resources. By biological characteristics,stability and the agronomic characters comparison research. Author preliminarlyselected high yield and good quality strains. Cordycepin was used as the mainparameter, differences of different strains were compared from these aspects suchas fruit body, solid culture medium, mycelia and fermentation culture. Based on theseresults, a dominant strain was screened. Moreover, then optimized rice-rice branmixed cultivation proportion. The main results were summerized as the following.1. Base on the ISSR molecular makers analysis the genetic relationships ofCordyceps militaris,11ISSR primers were screened out and chosen of identificationstrains,the total71clear bands were detected by PCR,64bands of them werepolymorphism bands, the average rate of diversity was90.1%. The UPGMA clusteranalysis results based on ISSR markers showed that30Cordyceps militaris strainscould be divided into8categories under similarity coefficient of0.74.When thesimilarity coefficient was detected at0.76, the third one could be divided intotwo subcategories. The results showed that the same area strains had the closergenetic relationship, the different regions strains had the distant geneticrelationships. There were large of genetic differences among Cordyceps militarissamples, genetic diversity was very rich. The diversity also showed geographicaldifferences in the distribution.2. Seven better strains were slected as better species by comparing the growthspeed and the fruit body yield.Comparison of Cordycepin among different strainsshowed that the strain containing the highest content of cordycepin is NO.10andNO.4.The contents of cordycepin were achieved by high-performance liquidchromatography (HPLC). At last, from the results of biomass and content ofcordycepin. It was concluded that NO.10was the better strain with high yield of fruit body and high content of active components.3. Results showed that when the rice and rice bran mixed proportion from50%to60%,fruit body have high yield, good quality, good economic and ecologicalbenefits.
