Study On Formulation And Preliminary Process Of Compound Cordyceps Militaris Granule

Cordyceps militaris (L. Fr) Link belongs to Ascomycota, Pyrenomycete, Sphaeriales, Clavicipitaceae and Cordyceps group. As functional component Cordycepin has an enhancement effect upon the immunologic function and is more in Cordyceps militaris than in Cordycrps sinensis, Cordyceps militaris can make it to be substitute of Cordycrps sinensis. But prior research shows that rats administrated by gastric perfusion with high dose become bad diarrhea. Aiming at the problem above, Attenuation of Diarrhea of Cordyceps was carried out. To establish a technical foundation for development of Compound Cordyceps militaris, application risk of Cordyceps is reduced on the research of extraction and compatibility of Cordyceps.First of all, best extraction condition is investigated. The experiment manifests that there is no significant difference between process of water extracting and alcohol extracting by determining content of Cordycpin. Then, based on the orthogonal design the optimal production process condition is temperature 80, 12 folds water for 4 times and 1 hour each time. Research on test of the maximal tolerance dose of extractive in mice shows that extractive still causes diarrhea. The effect of Cordyceps militaris extract on immunity is tested in mouse of clearance rate of charcoal particles and delayed type hypersensitivity, and the results show that phagocytic index a and weight difference between the same size of the two earflaps in original medicinal herbs group are significantly higher than those in the control group, but those in water extract group have no significant difference comparing with those in the control group. All these results support the notion that original medicinal herbs can increase the function of macrophage, induce delayed type hypersensitivity reactions of mouse and enhance cellular immune function, but the extract can’t.In order to improve safety and immunological function, try to mix with other herbs. Observation on diarrhea caused by Compatibility of Cordyceps and Grossularia burejensis Berger or Chinese yam, shows that Cordyceps compatible with the latter is safer than the former. Based on Intergrating Chinese and Western Medicine, Cordyceps, Chinese yam and coicis semen are used as raw material in prescription.After Determination of prescription, optimal process of Compound Cordyceps militaris Granule was studied. Chinese yam and coicis semen were extracted, concentrated, and made into dry powder, then mixed with Cordyceps, fructo-oligosaccharide and dextrin by ethanol.Content of polysaccharide in extraction of Chinese yam and coicis semen was analyzed, which is used in quality of control of intermediates. Cordyceps militaris and Cordycepin in Compound Cordyceps militaris Granule are identified by TLC, Cordycepin is determined by HPLC, which are used in quality control of Compound Cordyceps militaris Granule. The methods are simple,accurate,reproducible and can be used for the quality control of Compound Cordyceps militaris.Compound Cordyceps militaris is given 15g/kg by gastric perfusion twice in a day, and diarrhea rate in mouse is only 2.5%. Then Compound Cordyceps militaris without the extract of Chinese yam and coicis semen, in which the content of Cordyceps is the same with Compound Cordyceps militaris, is also given by gastric perfusion, but diarrhea rate is 27.5%. This research shows that the extract of Chinese yam and coicis semen can inhibit side effect of digestive tract induced by Cordyceps. All these research manifest that Compound Cordyceps militaris is safer than single herb Cordyceps.To verify immunologic function of Compound Cordyceps militaris, test of lymphocyte transformation, test of the macrophages of the abdominal cavity of mice phagocytose the erythrocytes of chickens, test of the activities of natural killer cell are carried out by gastric perfusion for 30 days, according to technical methods and requirements of determination of functional components of health food. The results show that the middle and high dosages have significant effects on all tests above compared with normal group, but low dosage has not. All of the studies manifest that Compound Cordyceps militaris has great effect on immunity in normal mice.All these research show that Cordyceps compatible with Chinese yam, coicis semen and FOS can inhibit adverse effects, and remain immunological function. And the preperation prcess and quality of granule are scientific and stable.
