The naturally distribution of Vaccinium uliginosum in northeast China, the Daxing’an and Xiaoxing’an mountain and Changbai mountain forest region, have a high decorative and medicinal values, but Vaccinium uliginosum is acidic plants, wild resources are limited, small fruit picking difficulty, can’t satisfy the needs of the market. In this study, with dormancy branches of Vaccinium uliginosum as test materials, and study the tissue regeneration rapid propagation and technical system, and based on the materials from micropropagation for the EMS mutagenesis method, alkali-tolerant mutant screening, germplasm resources for the protection, utilization and innovation lays the foundation,test results have the following:1The results indicated that the media for inducing shoot clusters was WPM+ZT2.0mg·L-1; the medium for subculture multiplication culture was modified MS+ZT0.5mg-L+IBA0.2mg·L-1,and the coefficient of multiplication was5.9after culture for30days;the medium for the rooting was1/4MS+IBA0.5mg·L-1with rooting rate of over94%with activated charcoal2g-L-1after culture for30days. Practices transplanting seedling was to tissue culture in sand20days, and then transplanting seedlings to the turfy soil:humus soil1:2, and survival rate o in nursery was up to78%.2The highest regeneration rate of78%,with12.6shoots per leaf,was obtained on the medium modified MS supplemented with2.0mg-L"1ZT and IBA0.3mg·L-1.ZTwas more effective thanTDZ、BA in inducing shoot regeneration from leaves of Vaccinium uliginosum;leaf differentiation ability was better than petiole and shoot tip.30.4%,0.4h EMS treatment would result in the coefficient of multiplication was3.6and survival rate reaches46.7%after culture for30days,when the0.1%EMS concentration ranged0.1%and the treatment duration was3days,the shoot tip of the coefficient of multiplication was1.8and survival rate reaches50%after culture for30days.4The Vaccinium uliginosum tissue seedlings resistance of critical concentration NaHCO3mutants for8mmg·L-1, will deal with half a fatal dose of EMS to the tube seedlings inoculated resistance in NaHCO3mutants appropriate dosage of medium, through the salt resistance acclimatizing medium and ordinary medium alternate training3times,2plants were alkali mutants.
