There is a dense and complex microbiota in bio-bed padding, which plays an important role in decomposing organic matter of animal feces and urine into gas, bacteria material and other nonprotein material. The present study was carried out to investigate the change of microbiota in bio-bed padding and feces of different states, animals (pig and chicken) and areas (ShenZhen and Nanjing). The paper included two trials.1. Studies on the similarity for bacterial communities in bio-bed padding and feces of pigMolecular methods PCR/DGGE based on the16S rRNA gene were used to study bacterial communities in bio-bed padding and feces of different pig pens (replacement gilt, lactating sow, weaning pig and finishing pig), different states (dry, wet, more and less) and different depth (10cm,15cm and30cm). The results showed, the bacteria configuration is abundant in bio-bed padding, indeed, mainly affected by the bacterial communities of feces. The bacterial communities in bio-bed padding which are same as feces take up60%, which four pens respectively is53-64%,56-67%,53-64%and57-68%. The number of bacterias in bio-bed padding were lager in the wet and more feces areas than the dry and less feces areas (P<0.01). The bacterial communities configuration of bio-bed padding and feces were varied in four pens. The similarity between feces and bio-bed padding respectively is30%,33%,36%and36%. Take the different depth as example, the bacterial communities are the most similar in the depth10cm and15cm, both more than50%.2. Studies on bacterial communities in bio-bed padding and feces of different source The comparison of bacterial communities structure in bio-bed padding and feces of different animal house (pig and chicken) and different areas (ShenZhen and Nanjing) based on16S rRNA gene method was conducted in the paper. The results showed, the bacterial communities in bio-bed padding which are same as feces take up79%in chicken house, the other21%from environment (feed, water and soil). There was a great variation on community structures in feces of pig pens in different areas, the similarity was less than30%. Between different animals, the similarity was37%in the community structures in bio-bed padding. It showed that bacterial communities configuration of bio-bed padding and feces were varied in pens of different animals and areas, which the factors of different animals had greater influence.
