The polymorphisms of GH(Growgh hormone)、 CAST(calpastatin)、 CAPN1(calpain)、CTSB(cathepsin B) and CTSF(cathepsin F) gene were detected by PCR-RFLP in304F2crossbredherd,(Limousin×Fuzhou)×Japanese Black Cattle, which commercially named Snow Dragon Black.The associations between SNPs and economic traits of the Snow Dragon Black Cattle were analyzed byusing least square of GLM procedure of SAS9.0. Then Real-time Quantitative PCR technology wasused to detect the expression of GH gene in12different genetype individuals, and analyze associationthe expression of GH gene between the economic traits. The purpose of this study is to find molecularmarkers which have a significant impact on the economic traits of the Snow Dragon Black Cattle. It willprovide scientific and theoretical basis for the new varieties of Snow Dragon Black Cattle.The mainresults were shown below:1、The polymorphisms of GH-P3, CAST-UOG, CAPN1316, CTSB-Aci I and CTSF-TaiⅠweregenotyped and associated analysis with economic traits of the Snow Dragon Black. The results were asfollows:(1) The GH-P3SNP was very significantly associated with chuck short rib (P <0.01).The AAgenotype of chuck short rib was0.42cm thicker than the AB genotype, and0.50cm thicker than the BBgenotype. The SNP was also significantly associated with the chuck flap and fat grade (P <0.05).The AAgenotype of chuck flap was0.52Kg weighter than AB genotype, and0.78Kg weighter than BB genotype.AA genotype of fat grade was0.95higher than AB genotype, and1.16higher than the BB genotype.(2)The CAST-UOG SNP was significantly associated with chuck short rib and fat thickness (P<0.05).The AA genotype of chuck short rib was0.32cm thicker than the AB genotype,and0.33cmthicker than the BB genotype; AA genotype of fat thickness was0.80cm thicker than the AB genotype,and0.47cm thicker than BB genotype.(3) CAPN1316SNP was no obvious association with economictraits studied.(4) CTSB-Aci I SNP was significantly associated with fat thickness (P <0.05), the BBgenotype of fat thickness was0.47cm thicker than the AA genotype, and0.37cm thicker than ABgenotype.(5) CTSF-Tai I was no obvious association with economic traits studied.2、According to the association analysis results,12Snow Dragon Black individuals which differentgenotypes of GH-P3were selected. Target gene expressions were detected in each muscle tissue usingReal-time Quantitative PCR System.The results showed that the gene expression of AA genotype wasthe highest, followed by AB genotype, and finally was BB genotype; And the expression of AAgenotype was significantly correlated with chuck short rib(P <0.05, r:0.9799), and BB genotype wassignificantly with meat color grade(P <0.05, r:0.9753).
