Tenderness is an important indicator of the meat. CAPN1is a member of calpain family, which participates in the muscle protein degradation process and is one of the important protein impacts on muscle tenderness. The CAST is the intracellular specificity inhibits the activity of CAPN1protein in cell. It affects the tenderness of the muscles with CAPN1. This study has scanned the genetic diversity of CAPN1and CAST genes among Lingshan chicken and Fast Partridge broiler chicken using PCR-SSCP, and analyzed the association the polymorphisms with meat quality. The main results were as follows:The genetic polymorphism of CAPN1gene on Exon5in the two breeds was detected. There’re two mutation points (C→T, G→A) had being identified. In this locus, He and PIC were between0.25and0.5, suggesting that there is moderate diversity in the two breeds. After X2test, Lingshan chicken has reached Hardy-Weinberg equilibrium and the Fast Partridge chicken did not. The heterozygosity and effective allele number of the two breeds are not high, and are geneticly identity.The genetic polymorphism of CAST gene on Exon3in the two breeds was detected, a A mutation point (C→T) had being identified. In this locus, Heterozygosity and effective number of alleles of the two breeds are not high, so the genetic identity.In this locus, He and PIC were between0.25and0.5, that there are moderate diversity in the two breeds of chicken population. The two groups of heterozygosity and effective allele number is not high, and geneticly identity.By PCR-SSCP and DNA sequencing, the polymorphism of CAPN1gene on Exon5was investigated, with two mutation points (C→T, G→A) with four different genotypes (AA, AB, AC and AD) had being identified. There was no significant difference of muscle fiber caliber between four genotypes in two breeds; the shearing force in AB genotype was very significantly higher than AD genotype in Lingshan males and Fast Partridge females; the shearing force in AA genotype was very significantly higher than AC genotype and AD genotype in Lingshan males; the intramuscular fat in AB genotype was very significantly higher than AA genotype and AD genotype in Fast Partridge females.By PCR-SSCP and DNA sequencing, the polymorphism of CAST gene on Exon3was investigated, with a mutation point (C→T) and three different genotypes (EE, EF, and FF) had being identified.
