Promoter is a switch to regulate gene expression. In the research of plant transge netic engineering, selection of promoter is very important, because promoters not onl y regulate expression intensities but also give gene specific expression pattern. With the deepening of plant transgenetic research, it needs more precise control about the h eterologous gene expression in transgenic acceptors, in order to avoid homologous dep endent gene silencing at the same, the application of chimeric promoters can meet the needs.CHS promoters from Petunia and Lily are flower specific promoters, they can specifically start gene expressing in flower tissue, but their expression ability are we ak. Enhancer which is divided into transcription and translation enhancer is a class of specific acting elements, they can improve the expression activity of promoters. In or der to explore the construction methods of promoters that have strong flower specific expression ability, this research fused35enhancer\OCSenhancer\Ω element with core r egion of Petunia flower specific promoter pPCHS and Lily flower specific promoter p LCH to become chimeric promoters. We want to analysis the effection of heterologou s enhancer on Petunia\Lily flower specific promoter activity through expression patter n of these chimeric promoters. Flowering results are obtained:1、Four unidirectional chimeric promoters that contain heterologous enhancer and core region of Petunia flower specific promoter\Lily flower specific promoter were constructed through overlap extension PCR.2、Plant expression vectors that contain four unidirectional chimeric promoters、c ontrol promoters and GUS report gene are constructed.3、Eight plant expression vectors were introduced into Torenia plant through Agr obacterium-mediated transformation,27to55transformed Torenia plants are obtained about each vector.4、GUS activity in leaf、stem、petal of transformed Torenia plants was detected by histochemical staining, the result shows that two unidirectional chimeric promoters contain35Senhancer and the two core flower specific promoters do not possess flowe rspecific expression pattern; two unidirectional chimeric promoters contain OCSenhance r and the two core flower specific promoters do possess flowerspecific expression patt er.5、Two bidirectional chimeric promoters that contain heterologous enhancer and c ore region of Petunia flower specific promoter\Lily flower specific promoter were c onstructed through overlap extension PCR.The above results indicates expression pattern changes of two flower specific pr omoters under the effect of different enhancers, it can give some suggestion about h ow to select strong flower specific promoters.
