A lot of good cultivated qualities which the wild rice contains but the cultivated rice lost during the long time period of evolution,such as anti-pest,stress tolerance,high tillering rate et al.Those qualities are crucial in keeping and improving the rice production.Wild rice was studied and applied intensively by researchers,and play an important roles in breaking the bottleneck of rice breeding.Based on the Chalin wild rice resource, its whole genome was transferred into rice cultivar R9810with pollen-tube pathway,after several generation’s selection we got different cultivars with good qualities as blast resistance,cold tolerance resistance,high tillering rate and the better cooking qualities.This thesis is combining the resource of Chalin wild rice,cultivated rice and the selected progenies to research their genetic diversity with RAPD. Selected progenies ys09203,ys09004,ys090015,ys090026and ys090027c numbered as1to5,which share apparently differences in gene and the5have the smallest diversity.Part of AOX non-coding region and some resistant genes located in chromosome11,12were found in analysis the polymorphic bands,and those sequence informations will contribute a lot to identification new genes.Considering those resistant phenotype in Selected progenies,we design various degenerate primers to research its genome and then have found25different resistant homologous gene sequences. With primer NL,the Pi2/9homologous gene were found in both of Chalin wild rice and Selected progenies.the homology ratios are98%,75%etc;with primer26LLR, the core domain of bacterial blight resistance gene Xa26homologous gene in rice was found,the homology ratio is91%;with primer BNBAR,a Bph14CDS homologous gene sequence was found,the homology ratio is91%. All of those homology sequences above have made a foundation for the further gene research and application of the Chalin wild rice.
