The Preliminary Reseach Of Tissue Culture About Three Kinds Of Mulberry In Hei Long Jiang Province

In this study,The primary culture,callus induction,subculture and rooting culture about mulberry was studied,the materials came form the winter buds, stems, leaves of good mulberry varieties" long Sang No.l" and the seeds of autumn mulberry and seedling mulberry. At the same time the tube seedings were used for refine seedling and transplanting test,the tissue culture system about three kinds of mulberry was established,at the same time reseach results will provide basic data for factory nursery of mulberry. The reseach results were as follows:1. The influence of the plant hormone6-BA, KT,NAA,2,4-D and other factors about mulberry primary cultureis was studied:The most optimum medium for the growth and differentiation of mulberry winter bud: MS+6-BA0.5mg·l-1+NAA0.2mg·l-1, the most optimum medium of mulberry stem segment The medium of WPM+6-BA1.0mg·l-1+NAA0.5mg·l-1was compatibly for induction of growth and differentiation of the axillary buds.The most suitable culture media for the seeds differentiation of mulberry seedlings and autumn mulberry:MS+6-BA1.0mg·l-1+NAA0.1mg·l-1.2. In subculture stage of mulberry seedlings, the different physicochemical factors were optimized on the effects of mulberry seedling increment, The best regeneration medium for three kinds of mulberry was MS medium that added0.1mg·l-16-BA and0.2mg·l-1NAA; Appropriate subculture cycle was about30days,in that case,mulberry seedling growed healthy and there was the highly regeneration rate of axillary bud.3. The suitable ratio of cytokinin6-BA and auxin NAA was helpful to improve the induction rate of callus. The callus of leaves and stems of the’ long sang No.l" was inducted, The best callus induction regeneration medium of leaves explant was MS added6-BA0.5mg-1-land NAA1.0mg-l-l,the best medium induction for callus from stem segment Was MS+6-BA0.2mg·l-l+NAA0.5mg·-l.The radicle of autumn mulberry and mulberry seedlings was selected as explants to induct the callus, MS+NAA1.0mg·l-1+6-BA0.2mg·l-1was suitable for callus formation.4.
