Abstract/Summary:
Cordyceps, one of the most valued traditional herbs in china. Nowdaysnumerouspharmacologicalactivitieshave been found in this medicinal fungus including immuno regulatory, antioxidantive antitumour, antiinflammation, antiatherosclerosis and so on. Until no w, Cordyceps can not grow in artificial conditions, but its asexual structre Hirsutella sinensishasbeen successed isolated from wild Cordycepsand produced by fermentation. Whether the fermented Hirsute lla sine nsis and natural Cordyceps have the similar chemical composition and medicinal value? It seems to be lackof enough research by now.This artical, we studied the antioxidant activity of Hrsutella sinensis water extract (HSE) from the chemical methods and PC12cells oxidative damage modelmethods1. FRAP method was applied to measure the antioxidative activity of Hirsutella sinensis mycelial water extracts. Results showed that the reducing power of lmg/mL water extract was equivalent to211.5±8.5μmolmLFe2+. Methodological study shows the reproducibility was3.8%, precision was3.1%; reliability correlation coefficient R2was0.9992; The recovery of this method were105.4±5.8%. The OD variation coefficient within30min afterreaction was3.0%.2. DPPH? radicals scavenging activity show when the HSE were0.5mg/mL,1.0mg/mL,2.0mg/mL,3.0mg/mL,4.0mg/mL,5.0mg/mL,6.0mg/mL, its activity we re6.6%±0.8%,16.1%±2.8%,31.6%±0.5%,45.9%±0.5%,60.0%±1.3%,72.0±1.4%,74.8%±1.0%.? OH scavenging activity also show when the HSE were2.0mg/mL,4.0mg/mL,6.0mg/mL, 8.0mg/mL、10.0mg/mL, its activity were27.5%±4.4%、43.6%±1.9%、50.8%±6.4%、60.9%±2.8%、64.2%±1.5%. When the HSE were2.0mg/mL、4.0mg/mL、6.0mg/mL、8.0mg/mL、10.0mg/mL,its ferrious iron chelatingactivity were22.0%±5.1%、44.8%±7.3%、53.6%±0.6%、62.2%±0.4%、66.4%±0.2%.3. we can see the water extract could improve the rate of survivalcells. When the water extract were5μg/mL,25μg/mL,100μg/mL, its rateof survival were41.1%±4.1%(P>0.05),45.2%±1.7%(P<0.05) and54.8%±3.3%(P<0.01), respectively, compared with the modle group36.5%±3.8%.The protective effect of the water extract on H2O2-inducedcytotoxicity determined by LDH assay. It show when the water extractwere5μg/mL,25μg/mL,100μg/mL, the activity of LDH was3000±450u/mL(P>0.05),2667±540u/mL(P>0.05) and2295±329u/mL(P<0.05), respectively, compared with the modle group2886±399u/mL.The level of SOD in cells which stands for the antioxidant ability hasalso been used in this research. When the water extract were5μgmL,25μg/mL and100μg/mL, the SOD activity were36%±9.9%(P>0.05),47%±17.2%(P>0.05) and63%±14.9%(P<0.05) compared with the modlegroup39%±12.1%.Further, we measured the ROS leval in cells throw DCFH-DA, resultsshowed the fluorescence intensity of the modle group and5μg/mL,25μg/mL,100μg/mL were316.21、353.21、293.96、248.59.All these results indicate that Cordyceps sinensis mycelium extracthas certain antioxidant activity, and the mechanism of protectionpossible relevant to its up-regulation of SOD, reduce ROS production.
