Exploring ecological relationship between the isolates and their host plants is based on understanding the composition and distribution of endophytic fungi, which is of great significance for the development and protection of medicinal resources. In this study, the unique Chinese medicinal plant Fortunearia sinensis was studied. To isolate endophytic fungi from health plant leaves, petioles and branches, the samples were collected from a F. sinensis natural community on Baohua mountain in Jiangsu province in the spring and fall of2011.1436strains in total were isolated from2700tissues and they were identified by morphological and molecular methods belonging to33genera. They are all ascomycetes involving ascomycetes6genera, hyphomycetes13genera and coelomycetes14genera. With the relative abundance>10%as index for differentiation, Alternaria, Fusarium and Pestalotiopsis were three dominant genera with the relative abundance of21.52%,19.64%and13.16%, respectively.The species cumulative calculation showed that the sampling number was adequate and effective for the diversity study of endophytic fungi from F. sinensis. From the variety of tissues, the Shannon-Wiener index and Margalef index of endophytic fungi in the petiole were respectively2.77and2.90which were larger than those in the laminas and twigs. And Endophytic fungi of F. sinensis in autumn with the Shannon-Wiener index of2.69and Margalef index of2.58were more diverse than in spring. The similarity coefficient between spring and fall was0.53, showing some differences of the community compositions. In different tissues, the similarity coefficient between laminas and petiole was the highest, which was0.77. The lowest coefficient was0.69between petiole and twig. The results mean that the community compositions of isolates exists the differences between the tissues less B than seasonal differences.F.sinensis leaves contain amount of ergenin and fortunearioside with some antioxidation activity. Based on the principle that endophytic fungi can produce the same as or similar to the secondary metabolites of their host plants, the isolates with high antioxidation activity were tried to explore in order to develop the resource of their natural antioxidants. With antioxidant activity as a target,60isolates were conducted in initial screen used T-AOC kit and one strain with higher activity was identified as Verticicladium trifidum. In our study, the reducing power (total antioxidant capacity) and radical scavenging abilities of its fermentation broth extracts were compared with rutin. The results showed that the extracts of the isolate had stronger antioxidant activity with good stability than rutin. According to the evaluation by O2-·/·OH/DPPH·scavenging in present study, O2-`and DPPH·scavenging activities of the extracts reached84.27%and84.25%, respectively. On the other hand, the scavenging rate against·OH was74.12%inferior to rutin at the concentration of1.0mg/mL.
