Rosa rugosa which has great ornamental and economic value is an important kind of ornamental shrub of genus Rosa, it is also the concernful parental in cross breeding. The study took2wild germplasms of Rosa. Rugosa(’HC’,’MP’) as female parents,2cultivars of Rosa rugosa (R. rugosa’Zizhi’,R. rugosa’Chongban’) and2cultivars of Rosa (R.’Red Hat’, R.’Pink Peace’) as male parents. This study researched the semi vitro culture of hybridized Rosa rugosa styles to explore the key factors which influenced the results of the semi vitro culture.The main results were as follows:1. Results of orthogonal test show that pH had significant effect to the length of pollen tubes through the incisions, but the influence of H3BO3was not significant; Sugar affected the percentage of styles with pollen tube significantly, but the influence of agar was not significant.2. After comprehensive analysis of the length of pollen tubes and percentage of styles with pollen tubes, we determined the optimized medium for semi vitro culture of Rosa rugosa styles after pollination:15%sucrose,0.005%H3BO3,0.1%agar, pH7.0.3. For the intraspecific hybridization combinations, when the pollen germination rate was greater than30%, the results of the semi vitro culture of style had no significant difference. For the interspecific hybridizatin combinations, when the pollen germination rate was greater than40%, there was no significant difference among the results of the semi vitro culture. To guarantee good pollination effect, the pollen germination rate must be greater than40%.4. For the intraspecific hybridization combinations and interspecific hybridization combinations, when we collected materials,4hours after pollination, the results of the semi vitro culture were best. It can help pollen grains to germinate on the stigma and enter into the style. when style was not cutted from plant.5. For the intraspecific hybridization combinations, there was no significant difference among the length of pollen tubes, as well as the percentage of styles with pollen tubes through the incisions.6. For interspecific hybridization combinations, the effect of style cutting length on the results of semi vitro culture of style was significant. There was a small amount of pollen tubes through the incisions of styles when cutting the upper one third-length style. When we cut the half-length style and full-length style, there were no pollen tubes.7. For the same hybridization combination, there was no significant difference between the flowers from the room water cultured flowering woods and field orchards as for the percentage style with pollen tubes through their incisions in all styles. For semi vitro culture of Rosa Rugosa style, we can use the flowers which were from the room water cultured flowering woods to extend the time span of collecting materials for our experiments.8. When we take the Semi vitro culture of hybridized Rosa rugosa style to study the growth situation of pollen tubes in the styles, the pollen germination rate must be greater than40%, We must cut the upper one third-length style when it reached4hours after pollination, and then used the optimum medium culture them for48hours with25℃, under dark condition, finally the length of pollen tubes could achieve the requirements of the study.
