Avian leukosis viruses (ALVs) are a group of retroviruses, which are divided intosubgroups A through J based on the antigencity of envelope protein gp85. But only subgroupsA, B, C, D, E and J were isolated in Chickens. Common clinical subgroups are A,B and J,while subgroups C and D are relatively rare in clinicals. Subgroup E Avian leukosis virusmainly exists in the form of the proviral in the host genomes. Subgroup J Avian leukosisvirus(ALV-J) was first isolated in1988by Britain Campton Animal Health Association. Inrecent years, ALV-J was isolated from commodity meat-type chickens, Egg type chickens anda flock of local breed. Also, ALV-J was isolated from clinical hemangioma cases. As early as100years ago, acute sarcoma had been found proved to be associated with ALV-A or byALV-B which was related with the cancer gene. The cell type is a kind of fiber cell tumor, butthere has no reports about acute fibrosarcoma induced by ALV-J associated virus. We got aALV-J from fibrosarcoma. The vieus can cause acute fibrosarcoma as helper virus in shorttime. So,we carried out the pathogenicity about it.1. Artificially created disease on acute chicken fibrosarcomas associated with ALV-JThe fibrosarcoma was collected from the neck of30-day-old817hybrid chicken inShandong in2010. In this study, the fibrosarcoma stored at-80℃was ground into a1:5suspension with saline, and then centrifugated in12000r/min for10min. The supernatant wasfiltered through0.22μm filter. Chickens was directly inoculated with the tumor suspensionfiltrate and supernatant from tumor extract-infected DF-1cell culture, respectively.2. Gross lesions of fibrosarcomas induced by inoculation with the filtrate of theoriginal tumor from the field caseThe actue fibrosarcoma was mainly attached to the subcutaneous of inoculation sites.When the sarcoma was cut, many fibrosarcoma nodules with different sizes could beSeen.3. Observation of histosection of the tumor sampleHistopathology indicated that the induced tumors were typical fibrosarcoma and shownsimilar histopathological changes. There were different cell types in different parts and even in the same field, there may be different types of cells in different areas of sarcoma. Somecells wre oval,spindle-shaped or irregular in shape, but most were long and slender fiber cellsat the ends. In the tumor, the fiber cells arranged in bundles, the nuclei had differnt shapesincluding prototype, oblong, irregular shapes and sizes. Some of the nucleus seems to besplitting.4. Inoculation chickens through different routes with tumor suspension filtrateOne-day-old817hybrid chickens were inoculated subcutaneously, musculely orintra-abdominally with filtered tumor extracts,0.2ml each. In the groups of inoculationsthrough the subcutaneous, in muscule and intra-abdominal, the tumor incidence was100%(20/20),95.2%(20/21),100%(20/20), respectively.5. Inoculation chickens through different types with tumor suspension filtrateOne-day-old Hyline brown chickens, SPF chickens,817hybrid chickens were inoculatedsubcutaneously, musculely or intra-abdominally with filtered tumor extracts,0.2ml each, thetumor incidence was78.3%,63.2%,85.0%,90%, respectively.6. Inoculation chickens in different doses with tumor suspension filtrateOne-day-old817hybrid chickens and SPF chickens were inoculated subcutaneously withfiltered tumor extracts,0.2ml each. The tumor incidence of817hybrids and SPF chickensinoculated with undiluted tumor extract filtrate was100%(10/10); When birds wereinoculated with tumor extracts in1:10and1:100, tumor rates were still in the range of80%(8/10) to100%(10/10) for both chicken lines; When birds were inoculated with tumorextracts in1:1000, tumor rates decreased to40%(4/10)or50%(5/10). There was no tumordeveloped in30days when birds were inoculated with the tumor extract of1:10000dilution.7.Inoculation One-day-old817hybrid chickens with with supernatant from tumorextract-infected DF-1cell culture.One-day-old817hybrid chickens were inoculated subcutaneously with filtered tumorextracts,0.2ml each. There were33.3%(2/6)chickens developed tumor mass when inoculatedwith supernatant from tumor extract-infected DF-1cell culture.
