In order to breeding out the varieties of good looking, strong resistance, fragrance, easy cultivation, this experiment of crossbreed from Cymbidium floribundum and Cymbidium faberiduse âDa Yi Pinâ in different species and interspecific was conducted. The studies mostly focused on the three aspects:The first was about the artifieial pollination and the growth of Cymbidium fruit. The basis research work of crossbreed of Cymbidium was carried out. The major objective was to find the potential obstacles in crossbreed of Cymbidium. The second was to establish the system of asylnbiotic propagation through the studies of asymbiotic germination of the crossbreed seeds and selection on appropriate culture medium. The three was to find the appropriate culture medium through design of different medium and concentrations of incretionăAC and juice on the callus formation and somatic embryogenesis of Cymbidium at the stage of the crossbreed rootstocks (protocorm) culture.The results show that:(1) The experiment about Hybridization was designed with4different types and each type had20flowers.68compages presented the phenomenon of ovary expand. If Cymbidium faberiduse as female parent crossbreeding in different species and inbred, the pollination fertility rate reached as high as100%. If Cymbidium floribundum as female parent crossbreeding in different species and inbred, the rate of pollination fertility was75%and65%.(2) The eapsule growing curve was single "S" Pattern. Different crosses had different best harvest time. If Cymbidium floribundum as female parent, the harvest time should prolong to110-130days after cross. If Cymbidium faberiduseâDa Yi Pinâas female parent, and Cymbidium floribundum as male parent, the harvest time was about90-150days after cross. The best appropriate inoculating time of inbred somatic embryogenesis of Cymbidium faberiduse was130-170days. It was effected by genetic and environment. We can get more protocorm if we chose the best time to harvest.(3) If Cymbidium floribundum as female parent, the best medium was Kc. If Cymbidium faberiduse as female parent, the best medium ZB11. Different medium has different rhizome bourgeon amount. The best culture medium which not only obtained many quantities generation, and had been short the work.So we should chose the best medium to get more cross-offspring.(4) The best proliferate medium for rhizome ofâCymbidium floribundumâxâCymbidium floribundumâis1/2MS+6-BA0.5mg/L+NAA0.1mg/L+CM20ml/L+AC0.5g/L.The best proliferate medium for rhizome ofâDa Yi PinâxâCymbidium floribundumâis MS+6-BA0.3mg/L+NAA0.5mg/L+CM20ml/L+AC0.5g/L and W+6-BA0.3mg/L+NAA0.1mg/L+CM20ml/L+AC0.5g/L. The best differentiate medium for rhizomeâDa Yi PinâxâDa Yi Pinâis1/2MS+6-BA1.0mg/L+NAA0.2mg/L+CM20ml/L, and the best rooting medium for rhizome is B5+6-BA1.0mg/L+NAA0.5mg/L+CM20ml/L.(6) The Pine bark, perlite and vesuvianite in the ratio of3to3to4were best media for temporary planting of tissue culture seedling of Cymbidium, and had higher survive rate and higher seedling growth rate than the other media. |
