Study On Cross Breeding And Aseptic Germination Of Embryo Of Cymbidium Hybridium

The research was based on 9 cultivars of Cymbidium hybridum and 1 cultivar of C.'Saint Raphin'(C. hybrid×C. sinense) imported from Korea and 1 cultivar of C. sinense imported from Guangdong by Taishan Forestry Academy.The most studies foeused on four aspects: The first was to probe intoe Cymbidium genetic background including the origon,the relationship of these imported cultivars.The second was about the artifieial pollination and the growth of cymbidium capsule.The major objeetive was to find the potential obstacles in hybridization of Cymbidium.The third was to establish the system of asylnbiotic propagation.The fourth was to research the effects of different medium and concentrations of incretion,CM,AC and banana juice on the callus formation and somatic embryogenesis of Cymbidium hybridum using an orthogonal design. The resultsshow that:(1)198 times Hybridization and 65 compages were designed among Hybrid Cymbidium Cultivars, C.'Saint Raphin'and C. sinense. 11 compages were still alive 4 month after the cross, 16.9% of all compages.Through aseptic germination 6 compages get offspring, 9.2% of all compages.The results showed that the mean fruit set of Hybrid Cymbidium×Hybrid Cymbidium , Hybrid Cymbidium×C.'Saint Raphin', Hybrid Cymbidium×C. sinense was 5.1% ,25% ,50%. The average germination rate reached 0% ,83.3%,71.4%of the three kinds of crosses by embryo culture.The rate of harvest mating was about 5.6%.Cross within cultivar harvest mating was 0%.Different crosses have different best harvest time. If C. sinense use as female parent, the harvest time should prolong to 180-210 day after cross, If C.'Saint Raphin'use as female parent, the harvest time is about 120-150 day after cross.we can get more PLBs if we chose the best time to harvest.(2)Through many cross experiment ,we filtrate several cultivars and compages who has better appetency. For instance, C. Lovely Moon'Gresent'×C.'Saint Raphin',C. Lovely Valley'Evergreen'×C.'Saint Raphin',C. Enzan Oberon'Gelee'×C.'Saint Raphin',C. sinense×C. White Crystal'Bigisse bidot',C. sinense×C. Lovely Moon'Gresent',C. sinense×C. Lovely Valley'Evergreen'. C. Lovely Moon'Gresent',C. Lovely Valley'Evergreen',C. Enzan Oberon'Gelee',C. White Crystal'Bigissebidot'were suit to be male parent, C.'Saint Raphin',C. sinense were suit to be female parent.(3)When C.'Saint Raphin'was use as female parent,the best medium was KC; When C. sinense was use as female parent,the best medium was MF. Different medium has different PLBs bourgeon amount.So we should chose the best medium to get more cross-offspring.(4) The best proliferate medium for PLBs is KC+NAA0.1 mg/L +6-BA0.5 mg/L+banana juice20g/L.The best differentiate medium for PLBs is KC+6-BA1.0mg/L+NAA0.5 mg/L + banana juice 15 g/L.The best proliferate medium for rhizome is VW+NAA0.5mg/L +6-BA1.0mg/L +CM20ml/L+AC1g/L.The best differentiate medium for rhizome is B5+NAA0.5 mg/L +6BA1.0 mg/L +CM20ml/L +AC3g/L.(5)Lichen were best media for temporary planting of tissue culture seedling of Cymbidium. Lichen has higher survive rate than the other media.
