Post-weaning swine diarrhea, with a relatively lower death rate and a higher morbidity rate,is a common problem on pig farms, seriously influencing pig growth, even causing great damage to pig-raising industry, is one of the most important factors that restrict the economy development of pig-raising industry. Based on the theory of Chinese veterinarian and the characteristic of the young animal, we think that the reason for the post-weaning swine diarrhea is Pi-deficiency, so, a kind of oral liquid, which is consitituded by Chinese herbs, against post-weaning swine diarrhea was study in our experiment.1Effect of different Chinese medicine compound on mice with Pi-deficiency syn-dromeTo obtain the optimal Chinese medicine compound on treating mice with Pi-deficiency syndrome(PDS),The experimental model of mice with PDS was induced by rhubarb decoction.Then the mice were treated with three different kinds of Chinese medicine compounds respectively. The time of cold resistance, feed conversion ratio and visceral index of mice were measured. As a result, compared with the PDS mice, the second compound showed a longer time of cold resistance, a higher feed consumption and visceral index. So we think that the second compound, which was constituded by Radix Astragali, Rhizoma Zingiberis, Rhizome Atractylodis Macrocephalae, Poria, possibly is the optimal Chinese medicine compound on treating the PDS mice.2Study on the preparing technology of the traditional Chinese medicine oral liquid against post-weaning swine diarrheaIn the aspect of preparing technology, the oral liquid was selected because post-weaning swine had a specificity of alimentary system and clinical need. A simple method based on ultraviolet spectrophotometer was developed for determining content of the polysaccharides in the aqueous extract. D-Glucose anhydrous showed a good linear relationship at the range of0~103μg/mL.The average recovery was104.1%and RSD=2.49%.Then, taking the content of polysaccharides as maker, we study the extraction times of the preparing technology. The result showed that the extraction process which extract the compound Chinese medicine by water, extracting3times, every1hours, adding5times amount of the water, can extract95%polysaccharides. To optimize the clarification process of the second compound aqueous extract with chitosan, the content of ploysaccharides was determined by phenol-sulfuric acid method with ultraviolet spectrophotometer, and the orthogonal design was used to study the three factors, including the mount of chitosan solution added, the flocculation temperature and time, taking the level of flocculation and the content of polysaccharides as markers. The optimal clarification procedure was A1B2C3. Under these conditions,the chitosan solution (1/5of the total volume)was added, the temperature of flocculation was20℃for24h. Compared with ethanol subsiding, chitosan flocculating method is feasible and economical, and the preparation shows a good clarity, it is expected to be applicable to the veterinarian clinic.3Quality standard of the traditional Chinese medicine oral liquid against post-weaning swine diarrheaTo establish the quality standard for the oral liquid, Radix Astragali and Rhizoma Zingiberis in this description were identified by TLC, and the content of polysaccharides which is the effective substance in the preparation was determined as the index by phenol-sulfuric acid method. The TLC results showed that the separation of the stains was apparent and the negative contrast was not disturbed, all of these proved that the method we used was specific. The phenol-sulfuric acid method which we used to determine the polysaccharides had high accuracy and precision. The quality control is feasible and can be applied to control the products from mass production. It can ensure the stability of the pharmaceutical quality. So we eatablish the content standard of polysaccharides which should be more than3.27mg/mL4Effect of the oral liquid against post-weaning diarrhea in different dosage on mice with Pi-deficiency syndromeTo explore mechanisms of the oral liquid in treatment of post-weaning swine diarrhea, the experimental model of mice with PDS was established by injection of resperpine,the mice were treated with the oral liquid in different dosage, respectively. The body weight, small intestinal propelling,visceral index and the content of serum amylase of mice were measured.The result showed that the oral liquid not only significantly regulates movement of small intestinal of mice with PDS (P<0.01), but significantly increases the thymus index and the content of serum amylase (P<0.01), in addition,improves the spleen index to a certain degree.
