Post-weaning swine diarrhea is a common problem in pig farms, seriously influencing piggrowth, even causing to die, causing great damage to pig-raising industry, is one of the mostimportant factors that restrict the economy development of pig-raising industry.In this study, according to the Chinese veterinary medicine theories and the etiopathogenisis,cardinal symptom and the pathopoiesis mechanism, twelve Chinese traditional medicines RadixPulsatillae, Rhizoma Coptidis, Cortex Phellodendri, Radix Scutellariae, Radix Astragali,RaidixPaeoniae Alba, Radix Angelicae Sinensis, Radi xAuc klandiae, Rhizoma AtractylodisMacrocephalae, Poria, Fructus Crataegi, Radix Glycytthizae were choosed to do the bacteriostasisexperiment in vitro. The used strains were E. coliO78, E. coliO20, E. coliO2, hog choleraSalmonella. The antibacterial activities of the Chinese traditional medicines against four strainswere tested by doubling dilution method in vitro, and the MICs were measured. Then five Chinesetraditional medicines which had better antibacterial activities were choosed to prepare threedifferent compounds, and also the MICs of the compound preparation were measured. Finally thebest compound was constituded by Rhizoma Coptidis, Radix Scutellariae, Radix Glycytthizae,Radix Aucklandiae, Fructus Mume.In the aspect of preparing technology, the oral liquid was selected united with pathogeny,clinicaled,dose etc. The extracting methods were selected according to the effective cmponents,pharmacological action, physical and chemical property etc. Rhizoma Coptidis were extracted byalcohol, the optimum extraction condition was investigated with orthogonal design method bydetermining the content of berberine. Radix Scutellariae was decocted by water, the optimumpreparing craft was gained by orthogonal test, taking the content of baicalin as study indexes; RadixGlycytthizae, Radix Aucklandiae and Fructus Mume were decocted by water, the optimumpreparing craft was gained by orthogonal test, taking the content of Glycyrrhizin as study indexes;The results showed that the oral liquid preparation technology was: Rhizoma Coptidis wereextracted by 70% alcohol, extracting 2 times, every 1.5 hours, adding 12 times amount of the70% alcohol. Radix Scutellariae was decocted by 16 times amount of water, extracting 2 times,every 1.5 hours. The others were decocted by 12 times amount of water, dipping 1 hour, extracting 2 times, every 2 hours. Then the ethanol was added into the concentrated liquid and the ethanoldensity in ethanol sedimention was 70%. The ethanol was reclaimed together with the alcohol ofRhizoma Coptidis. The oral Liquid was obtained with additive after the concentrated liquid wasdiluted by water.In the aspect of the quality standard, Rhizoma Coptidis, Radix Scutellariae, Radix Glycytthizaein the oral liquid against post-weaning swine diarrhea were identified by TLC; The main contentberberine Hydrochloride, baicalin, glycyrrhizin in the oral liquid were assayed by HPLC; The TLCresults showed that the separation of the stains was apparent and the negative contrast was notdisturbed. All of these proved that the method we used was specific; The HPLC results showed thatberberine Hydroehloride showed a good linear relationship at the range of 0.0032~0.016mg/ml; Theaverage recovery was 99.5% (n=6) and the RSD wasl.38%. Baicalin showed a good linearrelationship at the range of 0.0048~0.024mg/ml; The average recovery was 99.1% (n=6) and theRSD was 1.41%. Glycyrrhizinshowed a good linear relationship at the range of0.0024~0.012mg/ml; The average recovery was 99.75% (n=6) and the RSD was 1.12%.A systematic studies of the selection and prescription of the oral liquid against post-weaningswine diarrhea in terms of thepreparation, extracting, clarifying techniques and the quality standardwas finished for this oral liquid, which would be the basis of the next study of pharmacology andtoxicological action.
