Follicle formation and development is an important aspect of female mammal reproduction. Mammalian follicular development is a very complex process, paracrine, autocrine, juxtacrine, and endocrine factors and signaling pathways are essential for ovarian folliculogenesis. In recent years, has been reported that BMP-6play an important role in the rodent, ruminant and human folliculogenesis, the porcine as an important economic animals, BMP-6in porcine follicular is little know.In porcine mature follicles, Western Blot and immunofluorescence detected BMP-6protein-positive signal in oocytes and granulosa cells, indicating that BMP-6expression may affect folliculogenesis in the porcine ovary. In vitro oocytes maturation cultule in porcine, add the BMP-6protein in the cultuled medium, we found that adding BMP-6protein in the experimental group compared with the control group, and expel a significant increase in the number of oocytes of the first polar body, it suggesting that BMP-6can promote the in vitro maturation of porcine oocyte. Porcine ovarian granulosa cells were cultured in different concentrations of BMP-6in vitro, the results show that exogenous BMP-6protein can promote the proliferation of granulosa cells. Granulosa cells cultuled in different concentrations of BMP-6by72h, total RNA and protein was extracted, Real-time PCR and Western Blot analysis two key proteins of progesterone synthesis (StAR and3β-HSD) in the detection of mRNA and protein levels. Real-time PCR and Western Blot results showes that BMP-6in a dose-dependent manner to inhibit the expression of StAR and3P-HSD gene, suggesting that BMP-6can inhibit progesterone synthesis of granulosa cells. It suggested that BMP-6may promote the proliferation of porcine follicular granulosa cells and inhibition of granulosa cell progesterone synthesis to inhibit granulosa cell luteinization process, inhibition of follicular atresia, and to maintain the normal physiology of the follicle function.Porcine not only as an important economic animals, but also is a transgenic bioreactor field has broad application prospects of animal model. To understanding of BMP-6gene regulatory mechanism of porcine follicular development for improving the fertility of porcines, the more reasonable and effective development and utilization of a variety of resources, speed up the breeding of fine varieties have broad application prospects; at the same time provide a theoretical basis for understanding the role of BMPs family in ovarian physiology and developmental, in vitro maturation of oocytes a better simulation environment, and thus provide new ideas for scientific research and clinical diagnosis and treatment.
