The microstructure of normal retina and experimental retina in the Bufo gargarizansCantor were observed and described in detail.The experimental retina was the retina of Bufogargarizans Cantor after the optic nerve was damaged.0ur attention was paid to the optic systemin Bufo gargarizans Cantor. We made quantitative analysis of thickness of the normal retina,everynormal retinal layers,the experimental retinal and every retinal layers with sections stained by H.E.Meanwhile, the expression pattern of GFAP in the retinal-tectal system of Bufo gargarizansCantor during the regeneration of optic nerve was studied, using immunohistochemistry. Theresults were as follows:(1)The retina of Bufo gargarizans Cantor was similar with the retina of most vertebrates that therealso were ten layers. The ratio of the visual cells to the ganglion cells is about3.01:1,thecharacter of the retina belonged to the day animals, but in fact it is primarily a daytime latent,low-light environment of the night or rainy day activities.(2)The retinal structure in Bufo gargarizans Cantor was studied using electron microscope.Theresults were as follows:Pigment epithelial cells have a spherical phagosome observed undertransmission electron microscopy, also a large number of melanin granules, endoplasmic reticulum,fat droplets of submicroscopic structure in the distribution around the nucleus. Photoreceptor outersegment and inner segment can be clearly observed, which out segment body is divided into theinner segment and outer segment of the outer segment membrane disc a lot of intensive parallel tothe stack; close to the outer segment at the ellipsoid contains a large number of mitochondria;ellipsoid and inner nuclear layer exists between the myoid, myoid containing Golgi apparatus, endoplasmic reticulum, ribosomes. Observed in core layer to deep color of the Müller cell nucleus;ganglion cell layer observed in single-layer arrangement of the nucleus; nerve fiber layer can seecolumnar arrangement of fiber bundles. Scanning electron microscope can be clearly observedlayers of structure boundaries; two types of cone and rod-shaped photoreceptor cells, and theirsurface with longitudinal interlobular ditch.(3) Compare the contrast with the experiment, with the advance of injury time, the thickness of theretinal layers reduced, and then were increased and the end were decreased, basically the same asnormal retinal thickness after injury30d. The thickening of pigment layer, the outer nuclear layerand inner nuclear layer were up to the peak in7thdays after injury, and the maximum of otherlayers were in14thdays after injury.(4)The results of the immunohistochemistry demonstrate that GFAP expressed in IPL,Bufogargarizans Cantor normal retinal Müller cells where display the external membrane offilamentous GFAP staining, but the remaining retinal layers were no GFAP sex staining. Thenumber of Müller cells in the retina and the retina of the different injury periods increased first andthen reduced, and finally became normal. The peak appeared in3rddays after injury. The numberof injured Müller cells wre equal to the normal retinal Müller cell number in90thdays. Negativecontrol of the retinal layers no GFAP positive staining.
