Cashmere goats with its unique high-quality fur products become important economic animals, which has an important position in animals production. When the hair rapid growth, hair follicle cell division very quickly, and has high protein synthesis efficiency. Hair follicles to synthesize more protein every year, and has obviously year cyclical change.This study Comparison of Inner Mongolia Cashmere goat skin proteome in different periods, to obtain the structure and function of protein or gene which related hair follicles cycle or affects hair follicle development.This study establish the Inner Mongolia Cashmere goats form January to December skin protein SDS-PAGE electrophoresis maps, preliminary analysis of the skin protein expression. Inner Mongolia Cashmere goats skin protein SDS-PAGE electrophoresis maps obtain19major protein bands,molecular weight between189.38to14.91KD. The highest percentage content of total protein are bands of1,7,8,9,10,11, which the molecular weight were189.38KD,55.93KD,52.2KD,48.42KD,45.89KD,38.38KD respectively. During the protein sample in different periods of skin,the same bands percentage of total protein almost all keep in a similar scope, and no found some of a period of protein samples apparently high or low expression.Research using liquid nitrogen grinding method successfully separated and extraction of Inner Mongolia Cashmere goats skin protein, system optimization two-dimensional gel electrophoresis (2-DE) technology of Inner Mongolia Cashmere goats skin proteome, to effectively used in Inner Mongolia Cashmere goats skin2-DE maps construction.Using the above methods and techniques, succeed established Inner Mongolia Cashmere goats skin protein maps in different periods using pI3-10linear gradients. Research use bioinformatics technology to analysis the diversity proteins expressed in different periods.After the2-DE maps analysis, found that April and August appears relative more protein spots. The maps of different period skin protein has high similarity, and has higher matching rate between adjacent period. Protein analysis found that two protein spots only appeared in March2-DE map, two protein spot only found in July2-DE map and one protein spot only found in August2-DE map.Comparison of Inner Mongolia Cashmere goat skin proteome in different periods studies results show that in the selection of21abundant protein spots which coexpression in different periods, did not find them to keep a consistent change rule, but shows different protein spots in different periods expression of different law.Using one-dimensional electrophoresis technology and two-dimensional electrophoresis technology, comparison of Inner Mongolia Cashmere goat skin proteome in different periods, not only advantageous to understand the Inner Mongolia Cashmere goat skin protein expression and distribution, but also can provide important related data and information from the molecular level reveals the hair follicle cycle mechanism, thus lay the foundation to the further development and utilization of economic value of Inner Mongolia Cashmere goat.
