As an important part of meat consumption,goat meat has been attracted more and more attention by its consumers,including its meat flavor,nutritional value and meat classification.Physical properties and protein composition of different muscles are important for exploring meat quality and improving muscle quality.In this study,First we measured the composition of amino acids and fatty acids,analyzed the physical properties of triceps brachii,biceps femoris,longissimus dorsi,intercostal muscles and gluteal muscles;IDA method was used to build the database of total protein in five locations of skeletal muscle,then GO and KEGG analysis were performed;Based on the SWATH method,we analyzed the difference among the five parts of protein,and explored the function and pathway;Finally,the Western blot quantification and immunohistochemical localization of MYL3 protein were used to verify the results.The main results are as follows:1.After the significant analysis of 15 kinds of amino acids content in different parts,it was found that: The content of Glu,Asp,Lys,Arg,His in the longissimus muscle was significantly higher than that of the intercostal muscles(p<0.05);The content of Asp and His in the biceps femoris was significantly higher than that of the gluteal muscle(p<0.05).The content of Lys and His in brachii muscle is higher than that in intercostal muscles(p<0.05).2.34 kinds of fatty acids were measured in the muscles of triceps brachii,biceps femoris,and longissimus dorsi muscles respectively.After a significant analysis of the same fatty acid content in different parts,it was found that: The difference between methyl linolenic acid methyl ester(C18:2C9),methyl erucate(C22:1)and CIS-11,14,17-twenty-carbon three-acid methyl ester(C20:3N3)in different parts was very significant(P<0.01).3.1227 total proteins and 17915 peptides were identified in five parts of triceps brachii,biceps femoris,longissimus dorsi,intercostal muscles and gluteal muscles of Inner Mongolia cashmere goat musles sample,707 proteins and 8111 peptides were detected in the triceps brachii,727 proteins and 7899 peptides in the biceps femoris,647 proteins and 7282 peptides in the intercostal muscles,587 proteins and 6943 peptides in the longissimus dorsi,and 645 proteins and 7618 peptides in the gluteal muscles.4.The total protein of the five parts was analyzed by GO component enrichment.The result showed that those proteins enriched in 18 Biological Process,20 Cellular Componant and 20 Mollecular Function.Through the analysis of KEGG pathway,1098 proteins from 1227 total proteins were covered in 33 pathways.5.Through the analysis of SWATH data obtained by liquid chromatography mass spectrometry,166 differential proteins in Inner Mongolia cashmere goats were screened out,16 special expression proteins of triceps brachii,and 31 differentially expressed protein of biceps femoris were screened,and 49 special expression proteins of intercostal muscles,and 39 differentially expressed protein of longissimus dorsi were screened,and 31 special expression proteins of gluteal muscles.A total of 17 differentially expressed proteins related to muscle fiber were selected from five locations.4 differential proteins were screened related to muscle fiber structure including MYL3,MYL1,MYL6 B and MYH4.6.The difference in the expression of MYL3 in the triceps brachii,biceps femoris,longissimus dorsi,intercostal muscles and gluteal muscles of Inner Mongolia cashmere goats was detected by Western blot technique.The expression of MYL3 in biceps femoris was highest than in the other four parts,which was consistent with the expression trend of proteomics result.7.The immunohistochemical results showed that MYL3 protein had positive reaction in triceps brachii,biceps femoris,longissimus dorsi,intercostal muscles and gluteal muscles,but the expression in biceps femoris was obviously,which was in accordance with the proteomics and the Western blot results,and the localization results showed that MYL3 might be located on the type I muscle fiber.
