This experiment Arbas Cashmere goats mast cells in the positioning and the existence of tryptase, as well as the stationary phase of mast cells staining results, Arbas Cashmere goat production and disease diagnosis for the Ordos region provides a reference.Test is divided into three parts:the test Arbas Cashmere goats mast cells fixed in Carnoy’s fluid and4%neutral formalin (NBF) fixative for conventional staining (modified toluidine blue, the conventionaltoluidine blue, aldehyde fuchsin); test two Arbas Cashmere goats MC immunohistochemical staining using rabbit polyclonal antibody (32and immunohistochemical peroxjdase; test the general organization staining and immunohistochemistrycomparison of staining results.Obtained under the experimental conditions:1.This test confirmed that the mast cells are widely exists in different organs of Arbas Cashmere goats, mainly easy to reveal the site of environmental antigenic sites. Mucosal mast cells are distributed in the intestinal mucosa lamina propria and lung, and connective tissue mast cells are located in the connective tissue of the submucosal connective tissue and other tissues and organs.2.Carnoy’s solution can better reflect the number and morphological changes of MC in the tissues. NBF can significantly inhibit the cells stained. Carnoy’s solution is a more suitable for routine staining and immunohistochemical staining fixative.3.This experiment using MC tryptase polyclonal antibody and indirect immunoperoxidase Arbas Cashmere goats MC-specific positive reaction, confirmed the presence of tryptase.4. The immunohistochemical specificity, high sensitivity, accurate positioning, form and function.
