Jining gary goat was a species of lamb leathers in China, characterized by precosity and high productivity. The initial mating of female goats happened approximately 6 months old. The average yean percentage of primiparity was 163.1, general speaking, one year two fetus or two years three fetus, average yean rate is 293.7%. Mast cells were an important part of immune cells in goat uterus and oviduct, We researched the goat uterus mast cells of the staining method, We had a complete set of mast cells staining method. And the distribution, morphology and number changes of mast cells in goat uterus and oviduct during estrous cycle were observed. The histamine concentration in tissues of uterus and oviduct at different stages was also measured by modified fluorospectrophotometry, which provide the morphology materials for goat reproductive immunology so as to provide theoretical basis in enhance goat production and utilizating economic value.Experiment 1 Goat Uterus Mast Cells: Evaluation of Fixation and Staining TechniquesHistochemistry of goat uterus mast cells was characterized by histochemical methods using modified toluidine blue and alcian blue -safranin O in four different fixations. The results showed that alcian blue and modified toluidine blue gave specific staining of all mast cells populations in goat tissues that had been fixed in Carnoy's, but fixation in 10%NBF,4%paraformaldehyde and bouin fluid was unsatisfactory, mast cells were stained pooly by alcian blue and modified toluidine blue. Modified toluidine blue and alcian blue are good methods for mast cells of goat uterus. Experiment 2 The Variation of Mast Cells in Uterus and Oviduct of Goat during Estrous CycleThe distribution, morphology and number changes of mast cells in goat uterus and oviduct during estrous cycle were observed using modified toluidine blue (MTB) staining. The results suggested that the mast cell in uterus was mainly distributed in the myometrium, followed by the lamina propria, and sporadically scattered in the perimetrium. However, mast cells were not seen in the uterine caruncle and endometrium. In oviduct, mast cells were distributed in the mucosal plica, muscular layer and adventitia. Morphologically, round, oval or fusiform mast cells were present in all stages of estrous cycle. Significant degranulation in the mast cells was observed mainly during metaestrus. The amount of mast cells in the uterus and oviduct was in the order of metestrus > proestrus > diestrus > estrus. They were different significantly only in the myometrium(P<0.01), while there was no difference in lamina propria and oviduct. The results show that the number of mast cells in the uterus changes with the cycle of estrous change.Experiment 3 Ultrastructure was Compared for Mast Cells in the Uterine Horn of Goat During Estrus CycleObserve mast cells in the uterine horn of goat in different estrus cycle (proestrus, estrus, metestrus, diestrums) by TEM. The result suggested that there had sparse and vary in length of the microvilli in the surface of the mast cell in uterus horn. Nucleus lies in the central or the side of the central of the mast cell. Nucleolus can be observed and the chromatin thick distributing along the inside of the nuclear membrane. There were a large number of particles in intracytoplasmic which had different electron density. And the particles which had inconsistency size unevenly distributed. We also observed mitochondrium, golgi′s body and endoplasmic reticulum. Compared of four stage of estrus cycle, ultrastructure of mast cells was similar,but the particles in the cytoplasm had different electron density. Especially in the metestrus, the electron density of mast cell was low. As a result, the mast cell in estrus cycle had degranulation phenomenon, so they can secret mediators and cytokines.Experiment 4 The Concentration of Histamine in Uterus and Oviduct of Goat during Estrous CycleThe histamine concentration in tissues of uterus and oviduct at different stages was measured by modified fluorospectrophotometry. The results showed that the histamine concentration in the uterus and oviduct was in the order of metestrus > proestrus > diestrus > estrus. Histamine concentration in uterine horn has a significant changes during metestrus with other stages (P<0.05), while there was no difference in uterine, uterine cervix and oviduct(P>0.05). The results showed that the histamine concentration in the uterus horn changes with the cycle of estrous change.
