Liaoning cashmere goats are the very precious animal resources in our country,the cashmere which is produced by secondary follicles of cashmere goat has high economic benefit.MT is a very important factor to affect the growth of cashmere.MT can regulate the growth cycle of cashmere and change growth-related genes expression at the molecular level.Lnc RNA is an important regulators of eukaryotic gene expression,and participates in the development and differetitation of organism.But,lnc RNA had no detailed report in hair growth research.To explore the mechanism of MT affecting cashmere growth mediated by lnc RNA,we builded the four transcriptoms of melatonin-exposed Liaoning cashmere goat skin cells using RNA-Seq and excavated lnc RNA related MT in this study.Finally,data was analyzed using statistical methods.The main results are as follows:(1)We selected MT with three treatment conditions(1.0g/L 24h;0.2g/L 24h;0.2g/L 72h)and without any treatment of the Liaoning cashmere goat skin cells,and builded four transciptoms using RNA-Seq,named M1_24H,M2_24H,M2_72H and C.After the comparative anslysis of lnc RNA FPKM value in this four transcriptoms,we found a lot of differential expression lnc RNA.M1_24H has 32 differential expression lnc RNAs,among which 4 lnc RNAs up regulation and 28 lnc RNAs down regulation.M2_24H has 10 differential expression lnc RNAs,among which 4lnc RNAs up regulation and 6 lnc RNAs down regulation.M2_72H has 113 differential expression lnc RNAs,among which 5 lnc RNAs up regulation and 108 lnc RNAs down regulation.(2)Target genes of differential expression lnc RNA among M1_24H were enriched in negative regulation of transcription from RNA polymerase II promoter GO term.Target genes of differential expression lnc RNA among M2_24H were not enriched in GO term.Target genes of differential expression lnc RNA among M2_72H were predominantly enriched in cellular metabolic process biological_process,binding molecular_function GO terms.(3)Pathway analysis results showed that: Target genes of differential expression lnc RNA among M1_24H were enriched in Steroid biosynthesis.Target genes of differential expression lnc RNA among M1_24H were not enriched in pathway term.Target genes of differential expression lnc RNA among M2_72H were predominantly enriched in Cell cycle,Lysine degradation,DNA replication,and TNF,NF-κB signaling pathways involved in hair follicle development were found out in pathways which differential expression lnc RNA were enriched.Based on all the analysises,we confirmed that the treatment way of MT was the most beneficial to cashmere growth at 0.2g/L 72 h.By analysis of target genes enriched in NF-κB pathway,we found the target genes related lnc RNA respectively were XLOC_005914and XLOC_018763.Thus we confirmed the above two lnc RNAs molecules were associated with MT.(4)The results of q PCR assay showed that melatonin-related lnc RNA expression was increased significantly,and this result was consistent with the result of RNA-Seq.In conclusion,we reported MT related Lnc RNA XLOC_005914 and Lnc RNA XLOC_018763 for the first time based upon RNA-Seq and verified it by real-time PCR.Preliminary studies indicated that MT regulates the development of hair follicle and cashmere growth through the key lnc RNAs of Lnc RNA XLOC_005914 and Lnc RNA XLOC_018763.
