Cinnamomum longepaniculatum (Gamble) N. Chao is an endemic Lauraceae (Lauraceae) Cinnamomum (Cinnamomum) plant species in China. In recent years, so many studies have shown that volatile oil of Cinnamomum longepaniculatum leaves has efficacy on antibacteria. antiphlogosis, anti-cancer, desinsection and so on. It is an important raw material of perfume, medicine, industrial consumer goods and products of the chemical industry. But it is instable and highly volatile, and has a strong odor. To improve the stability of volatile oil of Cinnamomum longepaniculatum leaves and cover it’s odor,we studied the including preparation for volatile oil of Cinnamomum longepaniculatum leaves with β-cyclodextrin, and the stability of that. Meanwhile we observed the clinical efficacy of β-cyclodextrin inclusion compound with volatile oil of Cinnamomum longepaniculatum leaves on chicken naturally infected colibacillosis. The main results are as follows:1. Preparation of β-cyclodextrin inclusion compound with volatile oil of Cinnamomum longepaniculatum leaves:Two indexes including the ratio of obtained inclusion compound and inclusion rate of oil were used to evaluate the technologies based on orthogonal design, in which the proportion between β-cyclodextrin and volatile oil of Cinnamomum longepaniculatum leaves, β-cyclodextrin and water, the experiment temperature and time of stiring were considered as four main technology factors. Results:The optimal encapsulation conditions was ratio of volatile oil of Cinnamomum longepaniculatum leaves to β-cyclodextrin1:8(mL:g), ratio of β-cyclodextrin to water1:15(mL:mL), inclusion tempratuer40℃and inclusion time3h. The ratio of obtained inclusion compound was78.59%and the inclusion rate of oil was80.00%.2. Study on the formation and the stability of β-cyclodextrin inclusion compound with volatile oil of Cinnamomum longepaniculatum leaves:To confirm β-cyclodextrin inclusion compound with volatile oil of Cinnamomum longepaniculalum leaves by infrared spectrophotometry and TLC(thin-layer chromatography). The results shown that feature peaks of volatile oil disappeared or reduced in inclusion compound, but they still in the mixture; inclusion compound had the same chromatography points with volatile oil of Cinnamomum longepaniculatum leaves. It means that the compound prepared under the optimized condition was stable and had been included completely. Under condition of high temperature, high humidity and strong light, the content of oil in inclusion compound had no obvious change. For mixture, however, the content of oil was evidently decreased. It concluded that β-cyclodextrin inclusion compound with volatile oil of Cinnamomum longepaniculatum leaves was stable to light, heat and humidity, stability of inclusion compound was remarkably superior to mixture.3. Diagnosis on chicken naturally infected colibacillosis:Clinical examination and laboratory diagnosis on sick chicken. This chicken house had never been appeared deadly infectious diseases such as Newcastle disease, cholera fowl, but it had poor ventilation and the feed without disinfection. The sick chicken showed depressed, slow movement, dyspnea, claudication and defecated white or green stools. Results of dissection were that the pericardial cavity was filled with yellow liquid, the liver was stained yellow, intestinal distention and abnormal absorption of vitellose. To combined with epidemiological investigation, clinical examination and dissection results, we initially suspected to chicken infected colibacillosis or salmonellosis,or mixed infection. To make a definite diagnosis we made laboratory tests. All the results conformed to biochemical character of E.coli:smear microscopy of pathologic materials showed short rod-shaped gram-negative bacteria; pink colonies grown on MacConkey agar and colonies with green metallic sheen grown on eosin-methylene blue agar; the isolated bacteria could hydrolyze starch and grease, ferment glucose and lactose, methyl red test and indole test were both positive. Animal test showed that the isolated bacteria had pathogenicity. The clinical diagnosis and laboratory diagnosis indicated that the chicken had been infected colibacillosis.4. Observation on the clinical efficacy of β-cyclodextrin inclusion compound with volatile oil of Cinnamomum longepaniculatum leaves on chicken naturally infected colibacillosis:One hundred and fifty five-day-old chicken which were diagnosed as colibacillosis were randomly divided into5groups, every group had30chicken. Group I was untreated that received the basal diet, group Ⅱ, Ⅲ, Ⅳ and Ⅴ respectively received the basal diet supplemented with1%SMM (sulfamonomethoxine),7.2%,6%,4.8%β-cyclodextrin inclusion compound with volatile oil of Cinnamomum longepaniculatum leaves10days, other conditions were the same. Evaluating the efficacy of β-cyclodextrin inclusion compound with volatile oil of Cinnamomum longepaniculatum leaves on treating colibacillosis in chicken by recording the variation of clinical symptoms and mortality, and observing pathological sections. The results showed that:untreated group’s mortality was76.67%, mortalities of group II,III,Ⅳ and V were53.33%,43.33%,56.67%and63.33%in turn. Compared with untreated group, chicken in group II,III,IV,V were recoverd or colibacillosis was controlled after treating10days. The results demonstrated that:supplementation with β-cyclodextrin inclusion compound with volatile oil of Cinnamomum longepaniculatum leaves had the effects on treating colibacillosis in chicken, and7.2%volatile oil inclusion had the best efficacy.
