The cereal cyst nematode (CCN) is an important pathogen on wheat and other cerealcrops, it has serious impact on wheat production wheat food security.To know the distribution of cereal cyst nematode (CCN) in Anhui Province, Totally37cities and counties (districts) were surveyed. During heading stage of wheat,"Zig-Zag"method was used to collect soil simples for counting white female on the wheat roots. Theresults show CCN was detected from52%sample sites of Anhui Province wheat area;100%sample sites of northern part of Huai River wheat area, from7%sample sites of Jianghuaiwheat area, no CCN detected from wheat area of south of Changjiang river wheat area. Fivepoint sampling method were applied to obtain wheat rhizosphere soil samples in Wangxingvillage in Xiaoxian county, the risk of CCN and other soil-borne pathogen were assessed bythe South Australian Research and Development Institute. The results show that CCNdensity of wheat rhizosphere were reached the high risk value, which are consistent with thedetection of artificial density results and yield loss results.Cysts were separated by Fenwick&Oostenbrink method from the soil samples of wheatgrowing areas in the Huaiyuan, Fengtai, Lingbi, Jieshou and Yingshang, second-stagejuveniles (J2) were obtained by hatching cyst. CCN was identified based on morphologiccharacteristics of cyst perinea pattern and de-man data. The results showed that CCN inYingshang and other4counties were identified as Heterodera avenae, certain morphologicaldifferences exist among the5H.avenae populations and the difference was not significant, theYingshang population of H.avenae is more clearly distinguished from other4H.avenaepopulations.PCR amplification rDNA-ITS characteristic fragment of CCN from Huaiyuan, Fengtai,Lingbi,Jieshou and Yingshang population, downloaded H.avenae representative groups ofITS sequences of Zheng zhou, Henan Zhao path tree groups, Shandong single county groupsand Xuzhou, Jiangsu the Peixian groups from the Gen Bank database. build a phylogenetictree using the MEGA5.05together with Anhui groups. Found the genetic length of Huaiyuanpopulation is shorter than the other species, and the Huaiyuan population of H.avenae isdistantly related with other4H.avenae populations, but with no consistency to the geneticrelationships.Quadratic orthogonal rotation combination design method was used to analyze theimpact of temperature and time on the hatch number of wintering cysts indoor The results showed that the number of nematodes peaked at15℃to19days, the number of nematodes ata high level in the range of10~22℃and10~20d.Based on field observation on March, early symptoms in field of Maniu town, Datunvillage, Dulou village and Jiudian township in Xiaoxian coungy is obvious, Collectingsamples of wheat, Second-stage juveniles (J2) were separated from the root soil samples byshallow dish method, and J2in roots were detected by acid fuchsine stain medthod. J2isolated from the soil sample and roots were identified, J2in the root were found in Majintown, Dulou village and Datun.Resistant varieties experiments were carried on in Xiaoxian county, include Fanmai-5and other84wheat varieties were test resistant to H.avenae of Xiaoxian population. IncludeFanmai8, totally3high resistant wheat varieties were Screened. Include Luomai-8, totally16resistant wheat varieties were screened. Include Zhoumai-22, totally46middle-resistantwheat varieties were screened. Include Jimai-21, totally16susceptive wheat varieties werescreened. Include Zhoumai-18, totally4high susceptive wheat varieties were screened. noimmune varieties.
