Currently, arginine is an amino acid studied broadly, mostly concentrated in theeffect of arginine on immune or embryonic development, while it is few to combinethe two. In this study, we explore the effects of dietary supplemented with differentlevels of arginine on embryo development, implantation, newborn pups weaningsurvival from the perspective of the immune. We hope to improve the litter size andsurvival rate of pups from birth to weaning.Sixty estrus Rex Rabbits were randomly divided into five groups, three repeats,each repeat four rabbits. Rabbit breeded were fed supplemented with0(group1),0.30%(group2),0.45%(group3),0.60%(group4),0.75%(group5) L-arginine in thebasal diet. Determination interleukin-2(Interleukin-2, IL-2), interferon-γ(Interferon-γ, IFN-γ), interleukin prime-4(Interleukin-4, IL-4) of serum on day10,20,30of gestation. Determination NO of serum on the day0,10. The concentrationof immunoglobulin IgA, IgG, IgM and free arginine was measured in colostrum.Total rabbit born per litter, live farrowing rate per litter, weight of per litter wasrecorded. The weight, spleen, thymus of newborn pups were weighted. Theconcentration of IL-2, IFN-γin serum was measured. The IL-2and IFN-γ mRNA ofspleen relative expression levels were measured by fluorescence quantitative PCR.The pups weaning survival rate was recorded. The results show that: The serumconcentration of NO is significantly higher than group Ⅰ (P <0.05). Theconcentration of Th1-type cytokines IL-2and IFN-γin the group III~group Ⅴissignificantly lower than group Ⅰ (P <0.05) during the entire pregnancy. While, theconcentration of Th2-type cytokines IL-4in group II~Ⅴ compared with group Ⅰwas significantly higher (P <0.05). Total rabbit born per litter, live farrowing rate perlitter during group Ⅱ~Ⅴ was significantly higher than group Ⅰ (P <0.05), per litter weight of group Ⅲ~Ⅴ was significantly higher than the group Ⅰ~Ⅱ (P<0.05). There was no significant difference (P>0.05) during these thymus index andspleen index of the newborn pups in each group. The survival rate of group Ⅴ frombirth to weaning was significantly higher than group Ⅰ(P <0.05), however it wasn’tsignificant compared with groupⅡ、Ⅲ、Ⅳ(P>0.05). The concentration of IL-2innewborn rabbits was not significant difference each groups (P>0.05). The level ofIL-2mRNA expression in the group III~Ⅴ was higher than group Ⅰ~Ⅱ(P <0.05).The concentration of IFN-γgroup Ⅳ, Ⅴ was significantly higher than group Ⅰ(P <0.05), moreover it wasn’t significantly difference between group Ⅳ, Ⅴ andgroup II, III (P>0.05). The level of IFN-γ mRNA expression in group IV, Ⅴ wassignificant higher than group Ⅰ~Ⅲ(P <0.05). The study confirmed that thepregnancy Rex rabbit diet added certain L-arginine can change Th1/Th2cytokineconcentration;improving rabbits born per litter; strengthening the immunity ofnewborn rabbit to improve weaning survival rate.
