During the growth course of rice, weeds will contend with them for good living conditions, which will restrict the yield.Meanwhile it will have a bad effect on the quality of rice and add disease and pests transmission. Though the effective herbicide has contributed to protecting the rice with its extensive use, the herbicide residue has a very serious negative effect on rice, such as yield and quality reducing. The conventional breeding will take a long time, which is a little inefficient and there is not enough resistant resource. The breeds’ resistance can’t last very long time. So it is hard to reach the requirement. It breaks new ground in releasing new resistant breeds and enriching rice germplasm resource with the integration of efficient biologic technique and conventional breeding. In recent years, transgenetic technique is widely used in improving the crop resistance. EPSPS gene, which is resistant against glyphosate is more and more popular in breeding work than other resistant genes. EPSPS gene is the unique target of glyphosate. Glyphosate is a widely known herbicide, which will kill the crop randomly while it killing the weeds. The high expression of EPSPS shows a strong resistance against glyphosate. With the development of biology technique, producing ESPS gene with genetic engineering will be very significant in improving the yield and quality of rice.In the experiment, the author transferred the EPSPS gene from Brochypodium distachyon derived from Israel into the Jinongda838by Agrobacterium-mediated method, and obtained the plants which were resistant against glyphosate. The molecular detection had proved the existence and transmission of exogenous gene. The EPSPS gene was amplified by PCR and added the suitable restriction site. The EPSPS gene replaced the bar gene in the Pcambia3301, the carrier the gene expressed on. Then pCAMBIA33E1, the recombined carrier was established. By the Agrobacterium-mediated method, the target gene was transferred into rice and the resistant plants were obtained by antibiotic screening. It has proved that the EPSPS, the exogenous gene has been integrated into the genome of rice by PCR molecular detection. According to the result of RT-PCR detection, it showed that the target gene had transcribed successfully. And the transgenetic rice plants showed resistant against glyphosate by daubing glyphosate on surface of their leaves.
