R.. excise (Maxim.) Hara and R. japonica (Burm.f.) Hara var. glaucocalyx (Maxim.) Hara are perennial herbs which hole plant are used as medicinal materials with the function of clearing hot detoxify, anti-tumor and anti-virus eliminating inflammation. Domestic and foreign research focus on the chemical compositions, the extraction, identification and pharmacological effects, but little on the seed’s physiological and biochemical activity. This paper sampling analysed about the activity changes, storage methods after their seeds were harvested, the physiological and biochemical changes and restoring methods. And objective is to explore the reason of their seeds activity descend and the methods of extending the seed storage life.In the first year, the two seeds’germination potential, germination rate and vigor index all rose at the beginning and then declined during their seeds storage at room temperature. The collected seeds can truly mature after going through a latter stage of ripening.After the two kinds of seeds storage for12months with room temperature storage (15-25℃), low temperature storage (0-4℃), frozen storage (-20℃) and dry storage, the results showed that the effect with room temperature storage and dry storage were similar. The best was in4℃and the worst is in-20℃. Based on experimental research, the best way for long term preservation was with low temperature storage (0-4℃).After artificial accelerated aging, the seed’s germination potential, germination rate and vigor index declined with different degree, and have significant difference with control (p<0.05). With the analysis of variance, the recession of the seed’s vigor was in advance of that of their viability. With the longer aging time, the physical deterioration was deeper. The bearing storage ability of two kinds of seeds were both low. The bearing storage ability of R. excisa was better than that of R. japonica var. glaucocalyx.The analysis results of main ingredients of seven physiological indexes showed that the relative electric conductivity, SOD, POD, CAT, MDA, soluble sugar and soluble protein played a very important role in the process of seed’s aging, which were the main reasons of the seeds aging. However, seed aging was not only in physiology, biochemistry and form, but also in the genetic mechanism’s change. The aging reason and mechanism of seeds need further research.A series of analysis results showed that germination rate, SOD, POD, CAT activity, the content of soluble sugar and soluble protein of R. japonica var. glaucocalyx was better than that of R.excise.By restoring the aging72h seeds with three medicaments of PEG, KNO3and GA3, the vigor of seed was improved respectively. By orthogonal design, the results indicated that the effect of treatment with GA3was best for the two kinds of seeds. The condition of improving seed vigor for R.excise was as follow: soaking temperature for20℃, immerse concentration for250ppm, soaking time for36h. The optimum condition was that soaking time for24h, immerse concentration for250ppm, soaking temperature for20℃for R.japonica var. glaucocalyx.
