Asarum heterotropoides F.Schmidt var.mandshuricum(Maxim.)Kitag.is a perennial herb of Asarum L.which is the traditional material medica of Jilin province.It propagates by seed,but the seeds are recalcitrant,intolerant dry storage,not conducive to be stored and transported.In addition,when the seeds mature,the temperature of the northeastern region is very high,improper storage will lead seed deteriorate easily,those are the main constraints of production and germplasm of Asarum heterotropoides F.Schmidt var.mandshuricum(Maxim.)Kitag.In this paper,the physiological and biochemical characteristics of the seeds of Aspergillus nutrata were studied under different temperatures and water contents,the deterioration of the seed was studied.The suitable storage conditions of the seeds were determined by different initiators.The aging of the seed is suitable for the selection of suitable initiators for bad seed repairing,which is very significant to preserve Asiatica and germplasm resources.The main studies are as follows:1.Asarum heterotropoides F.Schmidt var.mandshuricum(Maxim.)Kitag.seeds were stored for 12months at different temperatures(room temperature,4℃and-18℃)and different seed water contents(50%,40%,30%,20%and 15%).With the storage duration,seed vigor was declining,but the seed vigor was different at different storage conditions.The rate of decline of the seed germination rate:room temperature>-18℃>4℃.The average germination rate was 57.26%at 4℃for 12 months’s storage.While the seeds were stored for 3 months at room temperature,the seeds lost its germination rate.The average germination rate of seed at-18℃for 12 months’s storage was 11.99%.The seed vigor at 30%water content was the slowest at the same temperature and at different water contents.The seed vigor at 4℃was the highest,and the germination rate was 87%,this was the best storage condition.2.During the Asarum heterotropoides F.Schmidt var.mandshuricum(Maxim.)Kitag.seeds storage,as the storage time went,the seed vigor was declining.With the increase of conductivity and MDA,soluble sugar and soluble protein contents decreased,and the activities of SOD,POD and CAT decreased continuously,the contents of GA3 and ZR decreased and the contents of IAA and ABA increased.At the same storage temperature,the conductivity and MDA of the seeds with different water contents were significantly increased with the storage time,but the conductivity and MDA of the seed with water content of 1530%increased slowly.The activities of SOD,POD and CAT decreased gradually,but the seeds of antioxidant enzymes at the water content of 1530%decreased slowly.the content of soluble sugar and soluble protein decreased gradually.The amount of endogenous hormones of the seed were little,GA3 and ZR decreased gradually.IAA and ABA increased gradually,But the content of endogenous hormones of the seeds with low water content were little.The order of change of physiological and biochemical indexes of the seeds was 4℃>-18℃>room temperature,with the same water content and temperature at different temperature.3.The results showed that there was significant positive correlation between the soluble sugar and seed vigor.The soluble protein,SOD,POD and CAT had very significant positive correlation with the seed vigor.Conductivity had very significant negative correlation with seed vigor.MDA and seed vigor had significantly negative correlation.GA3 and ZR were negatively correlated with seed vigor,IAA and ABA were negatively correlated with seed vigor,but none of them reached the significant level.4.Using 5 kinds of concentration of initiator to repair the seeds of deterioration,the experimental results showed that the effects of different concentrations of PEG-6000 for aging seeds of 1d、3d and 4d was better,the seed vigor of the average increased by 11.23%、36.75%and 25.00%.The effects of ASA on the aging seeds of 13d was the best,making the seed vigor of respectively increased by 16.66%,24.33%and 46.00%.
