And in recent years, the climate disasters, such as drought, become more and more frequentyl. It is verey important to raise the drought resistance of wheat and develop drought-resistant wheat varieties.Transcription factors also called trans-acting factor, which the function is the regulation of gene expression. Many stress resistance genes may be related to cold, drought and high salt stress-induced, or enhanced or decreased expression. Therefore transcription factors in plant resistance to adversity stress plays a very important role. OsAP22and RdreBl, the genes isolated from rice, which blong to EREBP/AP2class, were constructed into the vector YG8198and YK796and then transformed into wheat immature embryos by gold particle bombardment. And hope that the new crop varieties with high ability of wheat drought and salt resistance, stress tolerance breeding can be get by this way.The cell regeneration differentiation capacity of wheat is weak for wheat is monocotyledon. Thus, in contrast to rice, maize and other crops of agronomic importance, genetic transformation of wheat is still in the exploratory stage. Mature wheat embryos of Yangmai11,Yangmai12,Yangmai14,Yangmai15,Yangmai16,Yangmai17,Yangmai18,Yangmai19and which are important cultivar in Jiangsu province wereused as explant for culture in vitro. The pourpose of this study is to construct a high effective system of plantlet regeneration for mature wheat embrvos. The main results are as follows:In total,41To generation positive plants were obtained. Of them,8had produced offsprings.3of8lines in total were positive in T1generation using PCR assay at RNA level. The activity of SOD Significantly higher than the control. The Yangmai11, Yangmai12, Yangmai14, have excellent characters for both culture in vitor and agricultural train. So they are good acceptors for transgenic wheat. The method of mature wheat embryos culture in vitro found in this study can be reference for other researchers. The best cultivation condition for Yangmai14is KT concentration2mg/l, NAA concentration0.5mg/1. The chang of concentration of NAA and KT can not influence the differentiation rate of Yangmai15and Yangmai194.distinctly, but green points rate was notable effect.
