China Lacks of Agaricus bisporus germplasm resources, but the development of industrial needs strains of more resistance and disease resistance. The introduction of wild germplasm resources, especially native wild strains resources is one of the reliable ways to solve this problem.1picked a wild mushroom in the wet. lush lawn of orchard on September18,2010in Alar. Tarim Basin, which was roughly identified as the mushrooms are Agaricus (suspected wild Agaricus bisporus). This studies on the identification of suspected wild Agaricus bisporus, the biological characteristics on understanding its growth characteristics and ability to adapt to the existing cultivation conditions, and then the wild mushroom artificial domestication and cultivation. The main results were as follows:1. The suspected wild Agaricus bisporus was successfully identified wild Agaricus bisporus. Benzyl chloride method extracted mycelium genomic DNA of suspected wild Agaricus bisporus.and it was used suspected wild mushroom DNA as a template for PCR amplification with universal primers ITS1and ITS4.The results of amplification used1%agarose gel electrophoresis to detect. and amplified product was sent to Sangon Biotech (Shanghai) Co., Ltd.. Sequence was measured740bp fragments. ITS sequences of suspected wild Agaricus bisporus was compared with those registered in the DNA sequence database on Gen Bank, the identification results were obtained according to comparing other the homo logy sequence in DNA sequence database on Gen Bank. We got a sequence with accession number EF460354of Agaricus bisporus which ho mo logy was100%,91%coverage. Downloaded ITS sequences of Agaricus bisporus similar sequences and used DNAMAN software to do a diagram analysis. The combination of morphological characteristics for identification, comprehensive analysis and identification of wild Agaricus bisporus was identified wild Agaricus bisporus. This sequence uploaded to the Gen Bank nucleic acid sequence database, and1got the accession number JN807464.2. The biological characteristics of wild Agaricus mushroom mycelium was studied about four aspects of Carbon, nitrogen, temperature. pH. On the experiments of the carbon source, the results showed that wild Agaricus mushroom mycelium had differencet choice of carbon source, soluble starch was the most suitable carbon. On the experiments of the nitrogen source, the results showed that organic nitrogen was more conductive to the growth of wild Agaricus mushroom mycelium than the inorganic nitrogen. On the experiments of the Temperature, the results showed that the wild Agaricus mushroom mycelium could grow in the5-30℃and25℃is better than other temperature. On the experiments of the pH. the results showed that the wild Agaricus mushroom mycelium liked growth in a weak acid environment3. Fermented cottonseed hull plastic bag cultivation, successfully bred wild Agaricus bisporus. The screening of mother seed culture medium, original seed culture medium and culture materials were optimized. On the experiments of mother seed culture medium, the results showed that wild Agaricus bisporus leaching solution could stimulate the mushroom spore germination and were useful to growth of mushroom mycelium. On the experiments of original seed culture medium, grain medium plus the appropriate cow dung medium was the best to growth of the wild Agaricus bisporus mycelium. On the experiments of culture materials. Wild Agaricus mushroom mycelium in the fermentation of cottonseed hull culture materials could be normal growth, but mycelial growth was relatively slow, covered fungi package needed more than40days. From the way of cultivation, the plastic foam box cultivation, flower pot cultivation and massive casing cultivation could get fruitbody, but the mycelium kinked less, mushrooms knotted relatively late, parachuted easily, fruiting amount was verv low.
