Cotton stalk is a special stalk.It possess the general character of traditional stalk and special character of wood. So the microorganism.The purpose of this research is focus on the study of microbiological deterioration of cotton stalk. Isolation and identification of cellulose decomposing microorganisms from soil was composite with white rot fungi degradating lignin. We finally selected strains for degradating lignocelluloses of cotton stalk. Traditional harvest in the local cotton field possess the characteristic of5cm straw. Actual experiment of2/3cotton stalk. The purpose of the composite microbial strains collaboration theory is applied to screening on the degradation of contton stalk lignin and cellulose composite strain.The main results were as follows:Cellulose congo-red medium was used to isolate strains of cellulose-decomposing from soil in the study.According to the detection of CMC activity、FPA activity and the efficiency of the actual degradation,values5strains which were screened of cellulose-decomposing microorganisms are numbered T4,T3,T2,M4,M3. The experimental design of5temperature gradient are22、26、30、34、38℃.And5culture time of24、48、72、96、120h. The results showed that the culture time at72h and temperature at30℃was the best culture condition for cellulose-decomposing enzyme by the optimization of culture time and temperature value.Through of20day permentation period,every5days of determination followed by cellulose and lignin content.And at5,10,15,20days,the degradative ability of cellulose and lignin of cotton stalk was determined. At20days, the rates of degradation to cellulose were T220.91%、T321.43%、T422.19%、M416.74%、M319.74%; the rates of deradation to lignin were T211.12%、T311.46%、T411.22%、M412.08%、M312.73.The five cellulose bacteria mixed with Phanerochaete chrysosprium. Have three repeat and the pure culture of control,through of20day permentation period,every5days of determination followed by cellulose and lignin content. As shown in chart3-1and3-2,lignin and cellulose composite strains for the degradation of cotton stalk in general than the effect of degradation of a single strain. XT2+PC and XM3+PC combination is better.From the test,we can find the cellulose degradation and mixed bacteria in the effect of lignin,the degradation of mixed bacteria is better.By physiologicad and biochemical identification of bacteria, T2、M3、T4belonging to Cladosporium、Aspergillus and Penicillium.T3is homologous Bacillus axarquiensis、M4is homologous bacillus sp.
