We take7peony species and2wild species of peony as explants. Throughstoring and vigor determination of the5peony species of pollen grains, the bestconditions of storing peony pollen were screened; combined with breeding objectiveof species and variety among genetic relationship, florescence, color, fecundity and soon, In total6of peony and herbaceous hybrid material combination of parentsmatching, total design including distant hybridization between, in the four hybridcombinations, the average hybrid combination pollination130flowers, pollinationflowers were520flowers. The conventional hybrid with repeated pollination, pollenpollinated by the law, the pollination of fertilization with obstacles to the yukonhybrid combination for embryo save, get tissue and transplanting hybrid seedlings; Atthe same time for the hybrid seeds for sand hidden test and germination characteristicsresearch; Finally established combining general plant seeds, and for hybrid F1andinto the germination ratio of sprout rate for research. Test the main results are asfollows:1. Pollen storage and dynamic measurement: pollen vitality is kept a long time in coldstorage conditions (4℃); different variety of pollen in single varieties’ energy thestrongest vitality Dan; Different dynamic measuring method with10%sucrose liquidculture method more precise.2. Setting percentage of Hybrid seed: Setting percentage of Hybrid seed betweenpeonies was higher than the wild herbaceous peony. Setting percentage of P. suffruticosa ‘Fengdan’×P. suffruticosa ‘Danluyan’ hybrid seed is highest as88.25%,and the wild herbaceous peony and hybrid seed setting of P. suffruticosa‘Fengdan’×P. officinalis coll. is only33.13%, explain the distant hybridization between pooraffinity, strong influence.3. For hybrid young embryo differentiation impact: Basic medium MS has greatinfluence to the differentiation of embryo; Plantlets in vitro grow better; Thedifferentiation of100d embryo age is good; the highest of the hormone combinationdifferentiation rate is6-BA0.5mg·L-1, NAA0.2mg·L-1, culture medium is MS+6-BA0.5mg·L-1+NAA0.2mg· L-1+agar7g·L-1+sucrose30g·L-1(pH=5.8);Hybrid combinations of P. suffruticosa ‘Tianxiangduojin’×P. suffruticosa‘Huolianjindan’ is best and leaves born rate, differentiation rate was significantlyhigher than the other three combination,57.11%,34.67%.4. Rooting culture: Overall, with the age of embryos, rooting rate, average root length,and the number of average, root index were increased. And they were best when theage of embryos is100d. Rooting rate in combinations of P. suffruticosa‘Tianxiangduojin’×P. suffruticosa ‘Huolianjindan’(63.16%) is best. average rootlength, and the number of average, root index in combinations of P. suffruticosa‘Fengdan’×P. carthalinika East Georgia, lgoeti. is3.25cm,5.40,4.39. Result shows thatthe longer embryos age, the better rooting of hybrid plantlets in vitro and rooting rate ofdistant hybridization is lower than hybrid between varieties.5. Transplanting survival rate of plantlets in vitro: after transplant30d, statistics foundthat only100d embryos age P. suffruticosa ‘Tianxiangduojin’×P. suffruticosa‘Huolianjindan’ combination survived two trees. Transplanting the survival rate isextremely low.6. The influence of GA3on the F1rooting: rooting rate is not with GA3increasedconcentration of the rise. The effect of GA3concentration for100mg L-1treatmentis the best, the rooting rate is as high as77.27%.7.The emergence rate of F1generation: The emergence rate of P. suffruticosa‘Fengdan’×P. officinalis coll. is the highest,55.14%. Under GA3100mg L-1processing, combination of P. suffruticosa ‘Fengdan’×P. suffruticosa‘Danluyan’ is the highest,72.73%. Result shows that only through appropriate GA3solution concentration is beneficial to seedling emergence of hybrid.
