4tree peony cultivars and7peony cultivars were employed for this thesis. The vigor test was carried out on the pollen of4tree peony cultivars and4peony cultivars after storage, and the best condition for storage was obtained from the test results. In combination with genetic relationship between varieties, the parental selection of wide hybridized combination was performed on8samples of tree peony and peony.4kinds of wide hybridized combination, in which one had the average of264flowers pollinated, were selected finally. Therefore, the total of1057flowers was for pollination. The method of conventional pollination combined with repeated pollination and pollen mentor were used for hybridization. The florescence and mucus secretion of4peony cultivars which were female parents were observed. The primary analysis for the results of observation and statistics of abortion about wide hybridized combination was made. Additionally, have the impact of different concentrations of GA3on ’Fengdan’ hypocotyl elongation in vitro embryo, the main results of the test are as follows:1. Storage of tree peony pollen:the tree peony pollen had the best storage with a temperature of4℃;"Fengdan" of simple flower had the best performance on vigor among tested pollen with different varieties. The method of10%sucrose culture was more accurate than others of vigor test.2. Storage of peony:the peony pollen had the best storage with a temperature of—77℃; The results of pollen vigor tests with different varieties and the accuracies of different vigor tests were same as ones of storage of tree peony.3.Flowering habit of female parent and observation of mucus secretion:the initial bloom stage of4varieties was about3days; the full-bloom stage was the fourth day to sixth days after flowering; and the flower-fading stage was about2days. The period of mucus secretion of peony stigma was short and mainly concentrated in full-bloom stage.4. Hybridization:a total of226flowers of hybridized combination1were pollinated. Only42of them grew pods normally and produced73hybrid seeds; a total of175flowers of hybridized combination2were pollinated and had no flowers abnormally. Therefore, there were no hybrid seeds; a total of320flowers of hybridized combination3were pollinated. Only39of them grew pods normally and produced78hybrid seeds; a total of336flowers of hybridized combination4were pollinated. Only46of them grew pods normally and produced43hybrid seeds. It can be found that it was not easy to seed for wide hybridized combination due to some reasons.5. Effects for epicotyl elongation of "Fengdan" caused by various GA3concentrations:there was a best effect for epicotyl elongation of "Fengdan" when the GA3concentrations was200mg·L-1.6. Polyploid induction of mature embryo and bud of "Fengdan" influenced by various colchicine concentration:the provided test materials would be harmed by using the colchicine with high concentration. The polyploidy was umable to be found when tested using flow cytometer.
