The rabbit is increasingly subject to the attention of domestic and foreign markets, as a multi-purpose economic animals. With the rabbit breeding expansion of the scale,rabbit common diseases cause huge damage to the rabbit industry especially the rabbit diarrheal disease. Chemoprophylaxis and limit nutrition are used to control diarrheal diseases that effect is not satisfactory in aquacultural production. The study found that enteritis related to genetic correlation, and IL-23R plays an important role in Signal transduction of immune response.The first, we search the genetic variation of IL-23R’s CDS by sequencing.And then,analyze the impact of mutations on protein structure and function, screen candidate SNPs may affect the IL-23R function. Genotyping large number of samples by HRM technology(include rabbit of healthy114,enteritis181,low-fiber enteritis56). Experimental design based on the Case-control association analysis of susceptibility SNP and enteritis. Finally, we analyze that IL-23R genetic variation impact on expression level of IL-22which is a downstream cytokine of IL-23R.The results were as following:1. This analysis revealed that there were total18cSNP in coding sequence,10cSNP were synonymous, the other8cSNP were non-synonymous:c.254A>G p.Q85R,c.558A>T p.P186Q,c.809A>C p.L269F,c.854T>C p.L285P,c.944C>T p.P315L,c.1084C>A p.L3621,c.1475C>A p.R492N,c.1685G>A p.G562E, respectively. There are analysis identified that c.SNP245A>G of IL-23R is relevant to genetic resistance of digestive disorders by evolutionary analysis of coding SNPs, Multiple Sequence Alignment and protein domain prediction.2. Genotyping large number of samples by HRM technology (include rabbit of healthy114,enteritis181,low-fiber enteritis56). Experimental design based on the Case (enteritis)-control (controls of healthy), AA genotype frequency in the case and the control group (61.3%,69.7%), AA is the predominant genotype. Analyze risk factor for allele, allele A have harm for (OR=1.4330,95%confidence interval0.9813-2.0926, P=0.0619); Genotypes AA and GG enteritis has a overdominance protective effect(OR0.1830,95%confidence interval0.0893-0.3750, P<0.0001).3. The expression level of IL-22which was the sacculus rotundus of genetic resistance to low fiber diet type intestinal inflammation in Rabbits were measured, the results show that IL-22mRNA expression levels increased with the deepening of the degree of inflammation, The severe group was significantly higher than the normal group and minor group(0.01<P<0.05), the difference was not significant in minor group and normal group (P>0.05);IL-23R genotype AG, AA and GG that corresponds to IL-22mRNA expression levels was downward, but not significant (P>0.05)This is report to study the genetic polymorphism of IL-23R gene and association with incidence of digestive disorders in rabbits,can provide theoretical support for the follow-up study of IL-23R enteritis mechanism, and lay the foundation for breeding for disease resistance research.
