Wool produced traits are one of the important economic traits of wool rabbits and searchi ng the SNPs related to wool produced traits are important work in wool rabbits breeding.The wool rabbits that are jointly cultivated by Shandong Mengyin Yida Rabbit Industry Company and Shandong Agricultural University are used in this experiment.Eleven genes are selected as candidate genes including KRT31,KRT27,KRT85,SFRP2,Wnt10 A,PGR,IGF-1,IGFBP1,PROP1,FAIM and CDK2.The polymorphism detection of the 5' region is performed by pooling scquencing and a total of 21 SNPs were found SNPs were typed by MALDI-TOF technique and Wool Traits were measured.The as sociation analysis of SNPs and Wool Traits and the results are as follow:(1)In this experiment,a total of three sites were found to have a significant effect on wool production(P<0.05),which were the FAIM gene site-515,the PROP1 gene site-1020,and the CDK2 gene site-336.The position-515 of FAIM had an addictive effect on wool production.(2)There are eight sites were found to have a significant effect on wool length.The SFRP2 gene site-759,-1029,PGR gene site-1445,-1511,KRT31 gene site-524,KRT27 gene-865 had significantly effects on the length of villus(P<0.05).The KRT85 gene site-1010 had significantly effects on the coarse wool length(P<0.05).The SFRP2 gene-933 had significant effects on both villus and coarse wool length(P<0.05).The wool rabbit population were in Hardy-Weinberg imbalance at positions-524 and-865.The SFRP2(position-759 and-1029),PGR(position-1445and-1511)and KRT31(-524)had an addictive effect on the length of villus.The position-933 of SFRP2 showed an addictive effect ang dominance effect on villus and coarse wool length.The position-1010 of KRT85 showed an additive effect on coarse wool length.(3)We found five sites that had significant effect on wool diameter.The KRT85 gene site-1010,the FAIM gene site-515,and the IGF-1 gene site-1299 had a significant effect on villus diameter(P<0.05).The SFRP2 gene site-933 and KRT31 gene site-524 had a significant effect on the diameter of coarse wool fibers(P<0.05).The position-1010 of KRT85 and-515 of FAIM had an addictive effect on villus diameter respectively.The position-1299 of IGF-1 had bothdominance effect and addictive effect on villus diameter.The position-524 of KRT31 and-933 of SFRP2 had an addictive effect on the diameter of coarse wool fibers respectively.(4)Two sites were found to have a significant effect on the coefficient of variation of wool fiber diameter In this experiment.Among them,the position of PROP1 gene site-1020 had a significant effect on the coefficient of variation of villous fiber diameter(P<0.05).The KRT31 gene site-524 had a significant effect on the coefficient of variation of diameter of coarse wool fibers diameter(P<0.05).The position-524 of KRT31 showed both dominance effect and addictive effect on the coefficient of variation of diameter of coarse wool fibers diameter.(5)The haplotype construction of the SNPs of the target gene and associated analysis with Wool Traits revealed that: the three haplotypes of the CDK2 gene were significant differences in terms of wool yield,villus length,villous fiber diameter,and coefficient of variation of villous fiber diameter(P<0.05);The two haplotypes of SFRP2 gene showed significant differences in villus length and coefficient of variation of coarse wool fibers diameter(P<0.05);The two haplotypes of PGR gene had significant differences in wool yield and diameter of villous fiber(P<0.05),and the interactions of haplotype AC and TT had significant effects on the coefficient of variation of coarse fiber diameter and coarse fiber diameter(P<0.05).In summary,12 sites were found to have significant effects on Wool Traits.The haplotype and haplotype interactions on CDK2,SFRP2,and PGR genes have a significant effect on wool yield traits.The results showed that the KRT85,KRT31,KRT27,SFRP2,PGR,IGF-1,FAIM,CDK2,and PROP1 may be major genes related to Wool Traits in wool rabbits or associated with major genes.And these significant SNPs can be used as molecular markers for wool rabbit breeding.
