Antibacterial synergists for Sulfonamides belong to a class of diaminopyrimidine drugs by artificialsynthesis, diaveridine and trimethopriom are two most commonly used antibacterial synergists in China. These drugs can enhance the efficiency of Sulfonamides and Antibiotics, so they are widely used in veterinary clinic. With the unreasonable use or abuse of Sulfonamides, the residues of antibacterial synergists will inevitably occur. Long-term consumption of animal food products containing diaveridine and trimethopriom would be likely to cause harm to human health. Therefore, in order to strengthen the monitoring of these drugs, it is necessary to establish a method for simultaneous determination of diaveridine and trimethopriom residues in chicken tissues.Diaveridine and trimethopriom residues in chicken tissues were extracted with acetonitrile, followed by acetone, their extracts were defatted by hexane and then applied to positive ion exchanging MCX cartridge which was washed with0.1mol/L hydrochloric acid followed by methanol. The retained drugs were eluted with10%ammonium-acetonitrile. Elute was evaporated to dry, the dry residue was dissolved by the mobile phase and filtered. Diaveridine and trimethopriom were determined on C18column with ultraviolet absorption detection at270nm, using acetonitrile-water which includes0.01mol/L potassium dihydrogenphosphate (13/87, v/v) as the mobile phase, and quantified by external standard.Under these chromatography conditions established in the assay, the concentration of diaveridine and trimethopriom had a good linear coefficient relation (R2>0.999) in the range of0.05-4mg/L. When spiked diaveridine and trimethopriom concentration in chicken tissues were0.05mg/kg、1mg/kg、2mg/kg、4mg/kg, recoveries of diaveridine and trimethopriom in muscle、 kidney、liver and skin+fat tisssues ranged from73.63%to96.44%and from75.09%to98.72%respectively, the intra-day coefficient of variation (C.V) ranged from1.34%to7.18%and from1.59%to10.28%respectively, inter-day coefficient of variation (C.V) ranged1.26%to6.93%and from1.71%to6.28%respectively. The limit of detection(LOD) and the limit of quantification (LOQ) of diaveridine were0.02mg/kg and0.05mg/kg respectively; The limit of detection and the limit of quantification of trimethopriom were0.02mg/kg and0.05mg/kg respectively.The extraction and purification and detection methods of diaveridine and trimethopriom residues in chicken tissues established in this experiment apply to the determination of diaveridine and trimethopriom single or multiple drug residues in chicken tissues.
