To detect whether single intravenous emulsified isoflurane anesthesia affects antioxidantcapacity, liver and kidney function of miniature pigs, and determine the therapeutic index (TI) ofthe tail vein injection of the mice, to evaluate its intravenous application security, to explore a newway of miniature pigs anesthesia, at the same time provide an important reference for clinical trialsof the drug.“Glass bottle method”was used to compound different volume concentration of emulsifiedisoflurane. Induction of anesthesia was executed via miniature pigs ear vein, anesthesia wasmaintained with intravenous infusion of7%emulsified isoflurane1.6mL/kg/h via ear vein.Anesthesia was stoped80min later. To monitoring breathing, heart rate, body temperature, oxygensaturation, and noninvasive blood pressure respectively. Collecting cavity venous blood samples at0min,10min,20min,30min,45min,60min,80min,2h,6h,1d,2d,3d, and using the kit to detectchanges about antioxidant capacity and liver and kidney function.100mice were randomly dividedinto10groups, there are5male and5female in each group. ED50group (5groups) was injected0.5%-0.9%emulsified isoflurane according to the dose of0.014mL/g; LD50group (5groups)was injected0.9%-1.4%emulsified isoflurane at the same dose. The disappear of the rightingreflex proves the mice has been anesthetized, bliss method was used to calculate the LD50, ED50and TI.Animals had no response to noxious stimuli such as sound, acupuncture and clamp duringanesthesia; Breathing, heart rate, oxygen saturation, muscle relaxation and other vital signs werenormal; Awakening process was smooth, there was not nausea, vomiting and restlessness.Miniature pigs antioxidant capacity, liver and kidney function indicators changed duringanesthesia, but the difference was not statistically significant, and it was a physiologicalfluctuations within the normal range. The ED50 and LD50 of emulsified isofluraneis is83L/kgand167L/kg in mice. The TI is2.The experimental results showed that single intravenous emulsified isoflurane anesthesiadidn’t cause oxidative stress, and didn’t affect liver and kidney function, TI is low in mice, and itssecurity can be improved by controlled breathing. Emulsified isoflurane is expected to become anew drug of miniature pigs anesthesia, but it should need further toxicological studies to apply thedrug to the clinical.
