The Protective Effect Of Resveratrol On Arsenic-induced Organs Injury In Cats

Arsenic trioxide （As₂O₃） is a known environmental toxicant and potent chemotherapeuticagent. Significant correlation has been reported between arsenic exposure （including consumptionof arsenic-contaminated water and clinical use of As₂O₃） and dysfunction in different organs （brain,liver and lung）. In this study, we aimed to elucidate the effect of resveratrol with neuroprotectiveactivities on As₂O₃-induced oxidative damage and organs injury.Twenty-four healthy Chinese Dragon-Li cats of either sex were randomly divided into fourgroups: control （1mL/kg physiological saline）, As₂O₃（1mg/kg）, resveratrol （3mg/kg） and As₂O₃（1mg/kg）+resveratrol （3mg/kg）. As₂O₃+resveratrol-treated group were given resveratrol （3mg/kg）1h before As₂O₃（1mg/kg） administration.In present study, we determined the changes of （ALT, AST, total bilirubin, cholesterol,antioxidant enzymes, total arsenic, GSH and GSH/GSSG） in plasma and the changes of（antioxidant enzymes, total arsenic, GSH, ROS, MDA,8-OHdG） and histopathological studyPretreatment with resveratrol upregulated the activities of antioxidant enzymes, and attenuatedAs₂O₃-induced increases in reactive oxygen species and malondialdehyde production. In addition,resveratrol attenuated the As₂O₃-induced reduction in the ratio of reduced glutathione to oxidizedglutathione, and accumulation of arsenic in tissue and plasma. These findings suggested protectiveeffect of resveratrol on As₂O₃toxicity in cat and provide a better understanding of the mechanismwhereby resveratrol modulates As₂O₃-induced oxidative damage and a stronger rational for clinicaluse of resveratrol to protect organs against the toxicity of arsenic.
