objectThis dissertation aimed at observing the gonad toxicity of arsenic trioxide and exploring their possible mechanisms. It may offer the experimental and theoretical basis for the recognition of the long time damage and security estimate on the arsenic.MethodsIn this study, a chronic (continuous arsenic exposure for 20 weeks) exposure to arsenic model had been molded,in which different doses of arsenic (0.0μg/mL, 0.05μg/mL, 0.1μg/mL, 0.2μg/mL, 0.4μg/mL) had been administered in mice drinking wate. Any change of mice uterine, ovarian, breast and vaginal morphology and organization of ER, PR expression had been observed from the view of systematical morphology and chemistry angle of immune system. Arsenic trioxide's toxic effect to gonad and obstruction mechanism to incretion can be known and researched via gonad tectology, fix position examination in terms of sex hormone receptor, and the variation of serum hormone contents.Results(1) Mice exposed to different concentrations of arsenic in body weight of the gonads, showed morphological changes to varying degrees. And 0.20μg/mL, 0.40μg/mL group more pronounced, the larger its reproductive toxicity. Conclusion: The dose of arsenic trioxide in the experiment had the female gonadal toxicity.(2) ERα, PR appeared in mice exposed to different concentrations of arsenic in the uterine gland epithelial cells, stromal cells, ovarian granulosa cells of the cortex, luteal cells, ductal epithelial cells, vaginal epithelial cells, stromal cells and smooth muscle cells, etc. , and its level increased with doses, there is a growing trend.(3) Different concentrations of arsenic content exposed to mice showed that serum estradiol: 0.05μg/mL, 0.10μg/mL increased (P> 0.05), 0.20μg/mL Group 0.40μg/mL group with lower (P <0.05 ); serum progesterone level of 0.05μg/mL, 0.10μg/mL mice reduced 0.10μg/mL group and significant differences (P <0.01), 0.20μg/mL group and 0.40μg/mL group content increased ( P <0.05).This experiment testified that chronic arsenic exposure on female had obviously reproductive gland toxicity, and the toxin-producing mechanism is through the environmental estrogen of endocrine disruptors.
