Arsenic trioxide (As2C3) was used experimentally to treat and protect the Marek's disease of chicken, for observing the antagonistic effect of As2O3 on tumor resulted from vMDV and trying to find the way to treat and protect MD and other rumor, based on the antagonistic effect of As2C3 on tumor. Meanwhile some internal and external indexes were detected to expound the antagonistic mechanism of As203 on tumor. The external trial was on inducing effect on apoptosis of tumoral cell resulted from vMDV by As2O3. MDCC桵SB1 was cultured with different concentration of As2O3 for detected the antagonistic effect on MDCC桵SB1. The relative indexes were detected for expound the antagonistic mechanism of As2C>3 on tumor externally. The internal trial was on antagonistic effect of As203on tumor in clinic. 1 days chickens were infected by vMDV. The chickens were divided into 5 groups. The antagonistic effect of As2O3 on chickens with MD was observed in clinic by adding different dose of As2O3 hi daily feed. The relative indexes were detected for revealing the antagonistic mechanism of As2O3 on MD. The result showed that:It was testified for first time that As2O3 was active to induce the apoptosis of MDCC桵SB1 in external culture. The yellow-green chunk and pellet with dense fluorescence in nucleus and plasma of apoptosis cell of tumor was observed and some yellow-green broken segment was found with fluorescence microscope. The apoptosis body was found with local cell membrane protruding out Flow cytometry analysis showed mat there was a obvious diploid peak in front of Gl peak.It was found for first time that As2O3 was active to induce MDCC桵SB Ito produce IL-1, IL-2, IL-6 and TNF in external culture. This was the cause and mechanism of apoptosis occurred possibly.As2O3 was active to adjust the content of cytokines in plasma of SPF chickens with MD. The content of IL-1, IL-2 decreased, the content of IL-6 and TNF increased in plasma of chickens with MD. But by adding As2O3 in daily feed, the content of IL-1, EL-2 increased, thecontent of IL-6 and TNF decreased in plasma of SPF chickens with MD.As2O3 was active to adjust the antioxidant function of SPF chickens with MD. The antioxidant function of SPF chickens with MD debased, the activity of SOD, GSH-Px decreased and the content of MDA increased. But by adding As2Os in daily feed, The antioxidant function of SPF chickens with MD increased, the activity of SOD, GSH-Px increased and the content of MDA decreased. The result provided some theoretical basis for demonstrating the relationship between antioxidant system and the mechanism of MD as well as for expounding the effect of As2C>3 on antioxidant function.The content of free radiant and NO increased in SPF chickens with MD. Adding AsaOs in daily feed, the content of free radiant and NO increased in primary trial period, but then reduced after 24-36days. The results provided some theoretical basis for relationship between the content of free radiant and the mechanism of MD.The erythrocyte immune function decreased in SPF chickens with MD. But the function was enhanced by oral treat with AsjOs.
